| BackgroundGastric cancer is one of the three common malignant tumors.The discovery of biomarkers for monitoring the recurrence of gastric cancer and judging the prognosis of gastric cancer is the focus of current research.The coiled-coil domain containing(CCDC)is a protein supercoiled domain.CCDC77 is a member of the coiled-coil domain protein family,which contains two coiled-coil domains.By querying the HPA database,it was found that the expression of CCDC77 m RNA was down-regulated in gastric cancer tissues.The prognosis of gastric cancer patients in the low expression group of CCDC77 m RNA was worse than that in the high expression group of CCDC77 m RNA.It is speculated that CCDC77 may be related to the prognosis of gastric cancer.However,the relationship between the expression of CCDC77 and the prognosis and pathogenesis of gastric cancer patients remains unclear.ObjectiveTo explore the expression of CCDC77 in gastric cancer tissues,analyze the relationship between the expression of CCDC77 and clinicopathological characteristics,postoperative survival and postoperative recurrence of gastric cancer patients,and further study the overexpressed effects of CCDC77 on the biological functions of proliferation,migration and invasion capabilities of gastric cancer cells.MethodsThe relationship between the expression of CCDC77 m RNA in gastric cancer tissues and the prognosis of gastric cancer patients was analyzed by HPA database.By purchasing gastric cancer tissue chips and collecting tissue specimens from patients with the IIb-IIIc stage of gastric cancer after radical gastric cancer surgery in Luoyang Central Hospital,combined with the clinical pathological data of the patients,immunohistochemistry and western blotting were used to detect the expression of CCDC77 in gastric cancer tissues and adjacent tissues Analyze the relationship between the expression of CCDC77 and clinicopathological characteristics,survival and recurrence of gastric cancer patients,and further analyze whether CCDC77 can be an independent prognostic factor affecting survival and recurrence of gastric cancer patients.WST-1,wound healing and transwell invasion assays were used to detect the proliferation,migration and invasion ability of gastric cancer cell line HGC-27 and AGS.Results1.Tissue microarray results showed that the expression level of CCDC77 in gastric cancer tissues was lower than that in adjacent tissues,and the difference between the two groups was statistically significant.The expression of CCDC77 was not statistically different with different ages,genders,tumor size,and degree of differentiation,but was statistically significant with different TNM stages,tumor invasion depth and lymph node metastasis.The median OS and 1-year,3-year and 5-year overall survival rates of the high expression of CCDC77 group were higher than those of the low expression of CCDC77 group.Tumor differentiation,TNM stage and low expression of CCDC77 are independent risk factors that affect the survival of gastric cancer patients after surgery.2.The expression of CCDC77 in 76 cases of the stage IIb-IIIc gastric cancer tissues in our hospital showed that the difference in expression of CCDC77 between gastric cancer tissues and adjacent tissues was statistically significant.The difference between the expression of CCDC77 and different ages,genders,and tumor sizes was not statistically significant,but it was statistically significant with the differences between different lymph node metastasis and differentiation degrees.The median DFS and 1-year and 3-year disease-free survival rates of the high expression of CCDC77 group were higher than those of the low expression of CCDC77 group.Lymph node metastasis and low expression of CCDC77 are independent prognostic risk factors affecting postoperative recurrence of locally advanced gastric cancer patients.3.The WST-1 results showed that the cell proliferation ability of the overexpression plasmid transfection group of CCDC77 was significantly lower than that of the Vector group;the wound healing results suggested that the cell migration ability of the overexpression plasmid transfection group of CCDC77 was significantly weaker than that of the Vector group;the result of Transwell invasion assays is suggested that the cell invasion ability of the overexpression plasmid transfection group of CCDC77 was significantly lower than that of Vector group.ConclusionsCCDC77 is low expressed in gastric cancer tissues,and it is an independent risk factor that affects the survival and recurrence of gastric cancer patients.The overexpression of CCDC77 can inhibit the proliferation,migration and invasion of gastric cancer cell line HGC-27 and AGS. |
