Parkinson’s disease(PD)is an age-related neurodegenerative disease with a high incidence in the elderly population.The current global aging trend also makes the disease more stressful to society,so the research in the last decade has attracted considerable attention.At present,the treatment of Parkinson’s disease adopts the method of deep brain stimulation,but the improper use of deep brain stimulation will produce side effects on other neurons.It is necessary to analyze the parameters of deep stimulation.In addition,through the study of the pathogenesis of Parkinson’s disease,it is found that the abnormal basal ganglia activity is related to the main motor signs of Parkinson’s disease.Therefore,Izhikevich model and Hodgkin-Huxley model are used to simulate the structure of basal ganglia network.The effects of deep stimulation on basal ganglia network in early and advanced stage are analyzed respectively,Besides that,the effects of deep stimulation on Parkinson’s disease are also analyzed when considering direct and indirect pathways in basal ganglia network.The main research results of this paper are as follows:In chapter 3,four kinds of neurons in basal ganglia network are numerically simulated by Izhikevich model.The response ability of basal ganglia network is determined by analyzing the effectiveness percentage of thalamic response and observing the firing pattern of each neuron.The effects of the amplitude and period of deep brain stimulation on the network in early and advanced stage are analyzed.Studies have shown that deep brain stimulation in early stage only can affect the network when the amplitude is between 10 and 250,which can guarantee the normal response and normal dynamic characteristics of the network at the same time.If the amplitude is beyond this range,the overall dynamic characteristics of the network will be destroyed.In advanced stage,there is a higher sensitivity that a smaller amplitude can achieve the treatment effect.As for the effect of cycle,the study found that the cycle value of deep brain stimulation reached its peak at 150 in both mild and severe cases,but the overall trend was different.And the amplitude is different,the change of the cycle will have an effect on the firing pattern of other neurons.In chapter 4,the effects of direct and indirect pathways on Parkinson’s disease are investigated.Direct pathway and indirect pathway are adjusted respectively by establishing Hodgkin-Huxley model.When we adjust the direct path,the network response error rate fluctuates.The network will be at the worst when the value of direct path is-12.When the indirect path is adjusted,the error rate of network response will be declining.The state of the network does not change obviously in the process of adjusting the current difference value between the two pathways.When the current difference value changes,the closer two pathways are,the worse the state of the network will be.In addition,It’s found that DBS can effectively improve the network effect in proper interval by adjusting the amplitude and period of it,.
