Melanoma is a malignant tumor produced by the malignant transformation of melanocytes and is the main cause of death for all skin cancer patients.Resveratrol is a polyphenol compound with a wide range of pharmacological activities,such as anti-inflammatory,anti-oxidant,anti-tumor and anti-aging properties.The abnormal proliferation and metastasis of melanoma are affected by a variety of biological processes.SHC binding protein 1(SHCBP1)is involved in a variety of cellular processes and is related to the occurrence and development of many tumors.There are reports in the literature that resveratrol inhibits the proliferation of melanoma cells,but the molecular mechanism of resveratrol inhibits the proliferation of melanoma cells has not been fully resolved.This experiment is based on the results of previous studies found that resveratrol inhibits the expression level of SHCBP1 in mouse melanoma B16 cells,using CCK-8,flow cytometry,Western Blot,Ed U(5-Ethynyl-2’-deoxyuridine)staining Methods To elucidate the molecular mechanism of resveratrol in inhibiting mouse melanoma(B16)cells,and further clarify the biological role of SHCBP1 in B16 cells.The main experimental results obtained in this study are as follows:1.Resveratrol inhibits B16 cell proliferation,cell colony formation and cell migration.This effect is because resveratrol inhibits ERK and AKT signaling pathways in B16 cells,and promotes the expression of cell cycle inhibitors p21,p27,and p53 proteins.At the same time,it was found that resveratrol inhibited the expression of SHCBP1 protein in B16 cells.2.SHCBP1 has a higher background expression in B16 cells.By obtaining the B16 cell line that interferes with SHCBP1,using transcriptome sequencing to analyze the differential gene KEGG pathway,it is found that the differential gene is mainly enriched in cell proliferation,apoptosis,oxidation and inflammation-related pathways,indicating that SHCBP1 does affect the proliferation of B16 cells.3.Separately interfered with and overexpressed SHCBP1 in B16 cells,and found that SHCBP1 accelerated the G1/S phase process of B16 cells,promoted the phosphorylation of ERK and AKT,and further affected the expression levels of Cyclin D1 and P21,thereby regulating the process of cell proliferation.4.Under the conditions of SHCBP1 interference and overexpression,B16 cells were treated with resveratrol,and it was found that resveratrol inhibits the expression of SHCBP1,regulates the ERK signaling pathway and thus affects the cell proliferation of B16 cells.On the other hand,Resveratrol also regulates the proliferation of B16 cells through the AKT signaling pathway.Conclusion:This study clarified that resveratrol inhibits the proliferation and migration of mouse melanoma B16 cells by inhibiting the SHCBP1/ERK and AKT signaling pathways.To provide experimental support for the clinical treatment of resveratrol,SHCBP1 as a clinical diagnosis and targeted therapy for the treatment of melanoma provides a theoretical basis and experimental basis.
