| Objective:The purpose of this study is to explore the effect of preventive administration of Astragalus hawthorn on polycystic ovary syndrome(PCOS)induced by high-fat diet in rats,to study the effect of Astragalus hawthorn on insulin resistance,obesity and chronic inflammation of PCOS,and to further explore the multi-target effect of Astragalus hawthorN.From the perspective of traditional Chinese medicine,it provides a new perspective for the prevention of PCOS.Methods:High-fat diet was used to trigger obese polycystic ovary syndrome rat model.A random five group average distribution among fifty 3-week-old female SD rats leads to each group with 10 rats.The blank group rats were provided with and were offered the same volume of distilled water by gavage;the model group was provided with high-fat diet and were offered the same volume of distilled water as blank group rats by gavage.Astragalus group was fed with high fat feed and given Astragalus aqueous solution by gastric gavage.the hawthorn group was offered with high-fat feed,and given hawthorn aqueous solution for intragastric intervention;rats in Astragalus Hawthorn drug group were provided with high-fat diet and given the intervention of Astragalus Hawthorn aqueous solution by gavage.The duration of drug intervention was 15 weeks.Food intake and fasting body weight were recorded every week.From the 8th week all rats were observed by vaginal smear for 3 consecutive cycles.Ins,T,IL-6 and TNF-αwere gauged by ELISA after 8 weeks.After15 weeks of drug intervention,blood samples were collected and sacrificed after anesthesia.The fat of ovaries,uterus and genitalia was taken,and the wet weight was gauged to calculate the ovarian index,uterine index and perigenital fat index.One side of the ovary was fixed and the histology of ovary was observed.The levels of INS,T,IL-6 and TNF-αwere gauged by enzyme-linked immunosorbent assay.Results:1.Body weight and 24 h food intake:The body weight of rats in different groups would increase significantly over time(P=0.000).The results of comparison among groups showed that there were significant differences in the body weight of rats in different groups(P=0.000).As time went by increasely,the body weight of rats in model group was significantly higher than that in blank group,and the largest body weight in all groups is the rats in hawthorn,and the smallest body weight of rats is astragalus-hawthorn in the groups of administration.Analysis of 24 h food intake of rats With the increase of time,the 24 h food intake of blank group was the largest.From the overall change trend of the broken line after the fifth week,the 24 h food intake of high fat diet group in hawthorn group was higher than that of other high fat diet groups,while the 24 h food intake of astragalus and hawthorn medicine pair group was lower than that of all groups.2.Estrus cycle of rats:From the 8th week,vaginal smear observation was conducted for 3 consecutive cycles.The estrous cycle of blank group was stably around 4~5 days,and the preestrus,estrus,late estrus and interestrus appeared successively.the estrous cycle of model group was prolonged to 7~8 days,and the estrus cycle was disordered and the cell morphology was dysplasia.The estrus cycle was normal and the morphology and development of all kinds of cells were normal in Astragalus group and astragalus and hawthorn medicine pair group.Nearly half of the rats in the hawthorn group showed prolonged estrus cycle,which was in the late estrus and interestrus period for a long time,and the estrus cycle was disordered,and the cell morphology and development were irregular.3.Fasting insulin:experimental time affect the concentration of INS(P=0.006),and 8weeks between groups of rats INS level is significantly higher than 15 weeks INS levels in rats,and INS concentration there are significant differences between different groups(P=0.000)for eight weeks of the model group and hawthorn group of rats was significantly higher than astragalus membranaceus group and the drug group,significant differences(P<0.05).The INS concentration of rats in model group at 15 weeks was significantly higher than that in blank group.4.The concentration of T in blank group and astragalus group at 8 weeks was significantly lower than that in model group(P<0.05);The T concentration in model group and astragalus group at 15 weeks was still significantly higher than that in blank group(P<0.05),but the T concentration in hawthorn group and astragalus-astragalus pair medicine group was significantly lower than that in model group(P<0.05).5.The concentration of the inflammatory factors:with the increase of experimental time completely will not affect the concentration of IL-6(P>0.05),but between different groups of IL-6 concentration exists significant difference(P=0.000),8 weeks of rats,the blank group significantly lower than the model group,the hawthorn group(P<0.05),model group obviously higher than that of astragalus group,hawthorn,astragalus and hawthorn medicine pair group(P<0.05);At 15 weeks,the blank group was significantly lower than the model group,astragalus group and hawthorn group(P<0.05),and the model group was significantly higher than the astragalus group,hawthorn group and astragalus and hawthorn medicine pair group(P<0.05),and there was no significant difference between the two groups(P>0.05).With the increase of experimental time completely will not affect the concentration of TNF alpha(P>0.05),but different concentration of TNF alpha exist signifant differences between the groups(P=0.000),and 8 weeks each rat TNF alpha concentration,there is no significant difference(P>0.05),15 weeks between groups of rats significantly differences in the concentration of TNF alpha(P=0.000),15 weeks of blank group rats significantly lower than the model group,astragalus membranaceus group and astragalus and hawthorn medicine pair group(P<0.05),the model group was significantly higher than that of astragalus membranaceus group and astragalus and hawthorn medicine pair group(P<0.05).6.Organ weight index:the ovarian index of rats in blank group after 15 weeks was significantly higher than that in model group,astragalus group and astragalus and hawthorn medicine pair group(P<0.05),and astragalus group was significantly lower than that in model group(P<0.05);There was no significant difference in the uterine index among the groups at 15 weeks(P>0.05).Peripheral fat index of rats in blank group after 15 weeks was significantly lower than that in other groups(P<0.05),and the difference was statistically significant.7.Ovarian histological analysis:in the blank group,there were abundant follicles at all levels,with normal development,clear cell hierarchy,orderly and compact granular layer cells,about 7-8 layers,complete zona pellucida,and a small amount of follicles dilate-D;More follicles in the model group showed cystic dilation and loose connective tissue arrangement.The granulosa cell layer decreased by 2~3 layers,and the membrane cell layer thickened.The number of follicles at all levels in Astragalus group was sufficient and a few follicles were dilated.The number of follicles at all levels in hawthorn group was abundant,a few follicles were dilated,a few luteal granulosa cells were swollen,and the cytoplasm was loose and light stained.The number of follicles at all levels was abundant and a small amount of follicles were dilated.Conclusion:1.PCOS modeling induced by high fat diet:after 15 weeks of high fat diet induction,the concentration of testosterone in the model group was significantly higher than that in the model group;Model group showed disturbance of estrous cycle and immature development of vaginal epithelial cells.In the morphology of ovarian tissue,the follicles in the model group showed cystic dilatation,reduced granular layer and other PCO changes.The levels of IR,HA,inflammatory cytokines and obesity in model group were higher than those in blank group.2.Effects of Astragalus,Hawthorn and Astragalus-Hawthorn on PCOS in rats:Astragalus-Hawthorn has significant effects on improving the disturbance of estrus cycle,ovarian PCO,and high androgen level,glucose and lipid metabolism,high inflammation level and other aspects in PCOS rats;The effect of Astragalus membranaceus on PCOS rats was similar to that of Astragalus and Hawthorn,but Astragalus membranaceus had a poor effect on improving the high androgen level of PCOS rats.Hawthorn has a poor effect on the improvement of estrus cycle disorder,ovarian PCO and glucose and lipid metabolism in PCOS rats,but the use of hawthorn for a long time can reduce the high androgen level and high inflammation level,and other aspects of the effect is poor. |
