| Objective:To study the protective mechanism of Dangguibuxuetang on Blood Retinal Barrier(BRB)of Diabetic rats through the Müller cell pathway,and to provide laboratory basis for the clinical application of angelica tonifying Blood decoction in the treatment of Diabetic Retinopathy(DR).Methods:1.The expression of tumor necrosis factor(TNF-α)in retinal tissues of rats was detected by enzyme-linked immunosorbent assay.2.The apoptosis rate of Müller cells in rat retinal tissue was detected by TUNEL staining.3.The expression of Glu(Glutamate,Glu)in retinal tissues of rats was detected by enzyme-linked immunosorbent assay.4.The protein expression of glutamine synthetase(GS)in rat retinal tissue was detected by western blot.5.The expression of Kir4.1 protein in rat retinal tissue was detected by western blot.Results:Compared with blank group,the contents of TNF-α and Glu and the apoptosis rate of Müller cells in the retinal tissue of model group and experimental group were increased(P<0.05),while the protein expression levels of GS and Kir4.1 were decreased(P<0.05),the difference was statistically significant.Compared with model group,the contents of TNF-α and Glu and the apoptosis rate of Müller cells in all experimental groups were significantly decreased(P<0.05),the protein expression levels of GS and Kir4.1 were increased,and the high dose and medium dose groups in the experimental group were significantly increased(P<0.05),the difference was statistically significant.The results of TUNEL staining showed that each layer of retinal tissue was damaged obviously in the model group,while the retinal tissue in the experimental group was improved to vary degrees,with orderly arrangement and clear hierarchy.Conclusion:When DR occurs,the retina is in the state of ischemia and hypoxia,and the expression of TNF-α secreted by the retina is increased,the homeostasis of the internal environment is destroyed,the apoptosis of Müller cells is increased,the permeability of BRB is changed,the homeostasis of BRB is damaged,and the retina is damaged finally.It was found that the apoptosis rate of Müller cells was significantly reduced and the morphology of retina was improved after the treatment of DR rats by gavage of Angelica tonifying blood decoction,which indicated that Angelica tonifying blood decoction could protect BRB of diabetic rats through the pathway of Müller cells,providing laboratory basis for the clinical prevention and treatment of DR. |
