| Objective:In this project,we developed the Chinese medicine tea substitute Anheyiyang tea under the guidance of the theory of "Medicine and Food Homology ",and studied the effective ingredients and mechanism of action of Anheyiyang tea in the prevention and treatment of pneumonia by using Internet pharmacology technology.The immunomodulatory effects were further evaluated by in vitro experiments with the help of serum pharmacology and cell culture techniques,providing a basis for the clinical use and promotion of the herbal tea substitute Anheyiyang tea.Methods:1.To investigate the potential mechanism of action of Anheyiyang tea against pneumonia based on Internet pharmacology technology.Firstly,we used TCMSP database to screen the active ingredients of Anheyiyang tea,and used Gene Gards database to screen the predicted disease targets,construct the active ingredient-target network,and use STRING to construct the protein interaction network.2.Study on the immunomodulatory effect of Anheyiyang tea on mice based on serum pharmacology method.Forty Kunming mice,male and female,were randomly divided into four groups of 10 mice each,and gavaged with low,medium and high doses of Anheyiyang tea and equal doses of saline twice a day for 3,5 and 7 days,respectively.The MTT method was used to observe the effects of different doses and days of administration of Anheyiyang tea serum on the proliferation of splenic lymphocytes,and to determine the optimal doses and days of gavage.The blank serum group and experimental serum group were established,and the effect on the proliferation of splenic lymphocytes under different serum levels and different incubation times of the two groups was observed by MTT method.Blood was collected under aseptic conditions at 30 min,1 h,2 h and 3h after drug administration,and serum was isolated to detect the proliferative activity of T cells and IL-2 secretion activity in spleen.Results:1.The active ingredients in Anheyiyang tea are mainly quercetin,luteolin,kaempferol,beta-carotene,irisolidone,formononetin,nobiletin,licochalcone a,beta-sitosterol.Its core targets against pneumonia include IL-6,CRP,IL10,CXCL8,IFNG,CD40 LG,IL-1B,ALB,IL-4,MPO,TGFB1,CCL2,STAT3,MMP9,ICAM1,CXCL10,HMOX1,EGFR,SLPI,SERPINE1,NCF1,CXCL2,PPARG,TIMP1,VEGFA,IL-2,F3,CYP1A1,MMP1,IL-1A,PLAT,etc.2.The serum pharmacological results showed that the serum of all three dose groups promoted the proliferation of T lymphocytes compared with the blank group,and the medium dose group had the highest value-added conversion rate on splenic lymphocytes after7 days of intragastric administration(P < 0.05).At 48 h incubation,the 40% blank serum addition had the most significant effect on the proliferation of mouse spleen lymphocytes compared with the conventional culture.The drug-containing serum also had a significant promotion effect at 40% compared with the blank serum group.Serum in the 1h time phase promotes T cell proliferation and IL-2 production with the strongest effect.Conclusion:1.The process of Chinese herbal tea substitute Anheyiyang tea to prevent and treat pneumonia is accomplished through the joint action of multiple components and multiple targets.2.Aqueous decoction of Anheyiyang tea significantly promoted T-lymphocyte transformation response and IL-2 production,indicating that Anhe Yi Yang tea has immunomodulatory effects. |
