Study On Professor Xu HuiFu’s Treatment Of Pediatric Anorexia Based On Methods Of Data Mining And Network Pharmacology

ObjectiveTo use data mining technology to explore the rule of drug use of Prof.Xu Huifu’s prescription in the treatment of infantiles anorexia,summarize his experience in drug use,and analyze the possible drug targets and interactions of drug combinations with the highest confidence and high support and cluster analysis,so as to explore the mechanism of action of core drugs.MethodsA total of 335 children with anorexia treated by Professor Xu Huifu in the pediatric expert outpatient department of Wuhan Hospital of Traditional Chinese and Western Medicine from January 2019 to January 2021 were collected.A retrospective study was conducted on 162 cases of anorexia with dysfunction of spleen in transportation or qi deficiency of spleen and stomach according to the diagnostic criteria.The children’s name,registration number,gender,age,visit time,TCM diagnosis,classification,return visit,syndrome,prescription medication,etc.,were recorded in detail.Microsoft Excel 2019 software was used to count the frequency of traditional Chinese medicine.Apriori algorithm in SPSS Modeler 18.0 software was used to establish a correlation model for correlation analysis.SPSS Statistics 25.0 software was used for systematic cluster analysis.We try to find out the law of drug combination,summarize Professor Xu Huifu’s prescriptions for treating children with common witness anorexia and his academic thoughts.TCMSP,STRING,Metascape database and Cytoscape software were used to analyze the interaction relationship and related pathways in drug pairs and disease.Result1.Data mining of Professor Xu Hui Fu’s medication experience1.1 Common drug pairs and triplet drug pairsAccording to the systematic cluster analysis,Professor Xu Huifu’s common drug pairs are: grain bud--malt,lotus leaf– poria coco,rhizoma sinensis--Angelica dahurica,Endothelium comeum--hawthorn,yam--coix seed,and astragalus--paraffin.Triple drug use are: Atractylodes--licorice--poria cocos,Endothelium comeum--hawthorn--lotus leaf,grain bud--malt--magnolia officinalis,hawthorn--coix seed--white lentilfour combination drug use are: atractylodes--Atractylodes--morum kernel--licorice--poria cocos,malt--grain bud--magnolia officinalis--six Shenqu,Endothelium comeum--hawthorn--lotus leaf--atractylodes.1.2 Common core prescriptionsspleen loss of health certificate: grain bud,malt,magnolia officinalis,Six Divine Comedy,lotus leaf,poria cocos,licorice,hawthorn,Endothelium comeum,and Atractylostomy.Syndrome of Qi deficiency of spleen and stomach: Radix pseudostellariae,Poria cocos,Chinese yam,Atractylodes atractylodes,Coix seed,Amomum kernel,white lentil,licorice.1.3 Common addition and subtraction with the prescriptionWith nasal congestion,tears: xin yi,angelica dahurica;with abdominal pain: corydalis corydalis,etc.with night terrors,sleepless: add rushes,hook rattan;with recurrent respiratory tract infection,hyperhidrosis: paraffin,astragalus,floating wheat;red tongue,red pharynx and other thermal signs: Scutellaria scutellaria,wormwood;with dry stool: cooked rhubarb,magnolia,etc.2.Pharmacological analysis of the core drug networkThe main effective compounds of poria cox-lotus leaf in the treatment of infantile anorexia were quercetin,kaempferol,isorhamnetin and epaverine.The main targets were AKT1,IL6 and IL1 B.The main pathways are the regulation of the interaction between cytokines and cytokine receptors and the NF-κB signaling pathway.It can be seen that the drug "lotus leaf-poria cocos" through multi-target,multi-channel treatment of children anorexia.ConclusionIn this study,162 cases of clinical cases were retrospectively analyzed.Through statistical analysis,it was proved that Professor Xu Hui Fu used traditional Chinese medicine to treat infantile anorexia.Data mining method is used to extract medication rules,and Professor Xu Hui Fu’s treatment of infantile anorexia is summarized in detail,which can be summarized as "one Qi travel around,eliminating and tonifying simultaneously","if you want to tonify,the spleen should be transported before tonifying",and he is good at using sweet and warm products and wind medicine to treat infantile anorexia.At the same time,through network pharmacology,it was concluded that the mechanism of "lotus leaf-poria coco" in the treatment of infantile anorexia may be mainly through the interference of IL-6,IL-1β,Akt and other target genes,as well as the regulation of the interaction between cytokine and cytokine receptor and the signaling pathway such as NF-κB.
