| Objective:Using N-methyl-DL-aspartate(NMA)to construct a true precocious model of female rats,to observe the effects of Jiuwei Chushi Granule on the body mass,follicle stimulating hormone(FSH),luteinizing hormone(LH)and estradiol(E2)levels of hypothalamic GnRHmRNA and KISS-1 expression in female rats with central precocious puberty,to explore the mechanism of therapeutic precocious puberty,and lay the foundation for further application for hospital preparations.Methods:The SD clean grade immature adult rats were fed in a cage with two females and one male,and the female mice born within five days were selected after mating successfully.The points were divided into six cages,eight in each cage,and fed by the mother,weaned at twenty-two days of age,divided into cages with female rats for four days of adaptive feeding.Twenty-six days of SD female rats with body mass at forty six point four nine grams plus or minus eight grams were randomly divided into six groups in SPF laboratories:control group,precocious puberty model group,gonaririn group for injection,high dose group of Jiuwei Chushi granule,middle dose group of Jiuwei Chushi granule,and low dose group of Jiuwei Chushi granule,eight rats in each group.With the exception of the control group,the other five groups used N-methyl-DL-aspartate method to establish precocious animal models.Each of the control and model groups was given to one milliliter of saline,and group of high,medium and low doses of Jiuwei Chushi Granule were given Sixty-nine point four eight grams per kilogram,thirty-four point seven four grams per kilogram,and seventeen point three seven grams per kilogram respectively,and rats in western medicine group were injectioned gonadorelin in muscle one hundred microgrammes per kilogram.The body mass of all rats were measured every two days,the serum FSH、LH、E2 level was detected by enzyme linked immunosorbent assay,and the hypothalamus KISS-1、GnRHmRNA expression level of each group was detected by real-time fluorescence quantitative PCR method.Results:1.Vaginal opening time:compared with the control group,the vaginal opening in the model group was significantly earlier,the difference was statistically significant(P<0.05);compared with the model group,the traditional Chinese medicine group and western medicine group delayed vaginal opening,the difference was statistically significant(P<0.05);2.Body mass:compared with the control group,there was no significant difference in body mass in twenty-sixth day、twenty-ninth day、thirty-second day after birth(P>0.05),and the body mass of control group and model group was significantly higher than that in high dose group of traditional Chinese medicine in thirty-fifth day、thirty-eighth day,the difference was statistically significant(P<0.05);3.Organ coefficient:compared with the control group,the uterine coefficient of the model group was statistically significant(P<0.05);the uterine coefficient of the western medicine group and the middle dose group of traditional Chinese medicine group were all larger than that of the model group,the difference was statistically significant(P<0.05);the hypothalamic coefficient,ovarian coefficient and mammary gland coefficient of each group were not statistically significant(P<0.05);4.Sex hormone levels:Compared with the model group,the LH、FSH、E2 level of the control,western medicine,Jiuwei Chushi granule high and and medium dose groups were statissignificant(P<0.05),indicating that Jiuwei Chushi granule can reduce the level of LH、FSH、E2 in precocious rats;the difference of level in LH、FSH、E2 between low dose group,high and medium dose group(P<0.05),hinting that there is a quantitative and effect relationship in regulating sex hormone levels of LH、FSH、E2 in precocious rats;5.Uterine wall intima thickness and number of ovarian luteum:the endometrial thickness of model group was four hundred forty-four point one seven microns plus or minus sixty-two point seven five microns,significantly increased than that of model group about two hundred twenty-nine point three nine microns plus orminus ninety point five six microns(P<0.05),and the endometrial thickness of high and middle dose group was significantly lower than that of model group(P<0.05).The number of ovarian corpus luteum in model group was significantly increased(P<0.05);The number of corpus luteum of ovary in high and middle dose group and western medicine group was significantly lower than that in model group(P<0.05);6.Relative expression volume of GnRHmRNA and KISS-1 of the hypothalamus in each group:The relative expression of GnRHmRNA and KISS-1 of the hypothalamus in model groups was higher than that in the control group(P<0.05),the relative expression of GnRHmRNA and KISS-1 in the high dose group,low dose group and the Goenerilin group was lower than that in the model group(P<0.05),and the relative expression in the middle and low dose groups was statistically significant(P<0.05).The relative difference in the relative expression of GnRHmRNA and KISS-1 between groups,including high dose group,medium dose group,low dose group and western medicine group,is statisinsignificant(P>0.05).Conclusion:Jiuwei Chushi granule can inhibit the occurrence of sexual precocious puberty induced by N-methyl-DL-aspartate(NMA),reduce the level of LH、FSH、E2 in CPP rats,depress the thickness of e ndometrium and ovarian luteum production,lessen the expression level of KISS-1 and GnRHmRNA of the hypothalamus in central pre cocious puberty rats,Jiuwei Chushi granule may play the role of inhibiting central precocious puberty by regulating hypothalami c-pituitary-gonal axis. |
