Polygonum cuspidatum is the dry rhizome and root of Polygonum cuspidatum Sieb.et Zucc.It has the effects of relieving dampness and relieving yellow,clearing away heat and toxic substances,dispelling blood stasis and relieving pain,relieving cough and resolving phlegm.It is used for damp-heat jaundice,drenching turbidity,entrapment,rheumatic arthralgia,carbuncle swelling and sore poison,water and fire burns,amenorrhea,syndrome,bruises,lung-heat cough.At present,the common processing method of Polygonum cuspidatum decoction pieces is to soften Polygonum cuspidatum medicinal materials and then cut them.Fresh Polygonum cuspidatum medicinal materials have a high water content,and it takes a long time to dry on natural conditions,and may cause mildew during the drying process;Polygonum cuspidatum medicinal materials are The water softening process may cause the loss of active ingredients.There is also a processing method of fresh slices at the origin,but the key processing parameters involved are not stipulated,and the above-mentioned factors can affect the quality of medicinal materials.Based on the above-mentioned problems,this thesis takes Polygonum cuspidatum as the research object to study the key technologies of the integration of production area processing and processing.Through the optimization of the integrated production technology of Polygonum cuspidatum decoction pieces,the comparative study is compared with traditionally processed Polygonum cuspidatum decoction pieces,and two types of decoction pieces are analyzed.The similarities and differences between the quality and efficacy of the drug.1.Research on Integrated Processing Technology of Polygonum CuspidatumCombining multi-index comprehensive scoring method,single factor experiment,orthogonal experiment design and other methods,the methods and techniques of net preparation,cutting and drying of Polygonum cuspidatum were optimized.The final integrated process is: take fresh Polygonum cuspidatum medicinal materials,wash Polygonum cuspidatum with 18 times the amount of water,wash 3 times,rotate speed 45r/min,and wash 30 s each time.After washing,dried Polygonum cuspidatum until the moisture content is about 35%-45% and cut into it.After cutting,use 60℃drying method to dry,the drying time is 7h,the paving thickness is 3cm,and it is turned once during the drying process.2.A comparative study on the quality of integrated Polygonum cuspidatum and traditional decoction piecesWith reference to the contents of the Pharmacopoeia,the properties,color,moisture,ash,extract and content determination of the two processing methods were tested.In addition to the two components specified in the Pharmacopoeia,resveratrol and emodin-8 were added.-O-β-D-glucoside and emodin methyl ether content determination,a comprehensive comparative study of the two processes of decoction pieces,the results show:(1)The properties of the two types of decoction pieces are in compliance with the pharmacopoeia,and the traditional decoction pieces have better color.The integrated decoction pieces are slightly deeper;(2)The inspection items and identification items of the two types of decoction pieces are in compliance with the pharmacopoeia;(3)There are certain differences in the content of five components in the decoction pieces produced by the two different methods.The content of emodin-8-O-β-D-glucoside is higher than that of traditional decoction pieces,while the content of resveratrol,emodin and emodin methyl ether is higher in traditional decoction pieces,and polydatin and resveratrol are in The total content of Polygonum cuspidatum integrated decoction pieces is significantly higher than that of traditional decoction pieces.The fingerprints of the integrated decoction pieces of Polygonum cuspidatum and the traditional decoction pieces were compared by HPLC.Among them,the fingerprints of the integrated pieces of Polygonum cuspidatum were matched to 17 common peaks,the traditional pieces of Polygonum cuspidatum were matched to 13 common peaks,and the two types of decoction pieces were matched to 11 common peaks.There are some differences in the fingerprints of the two kinds of decoction pieces.3.In the experiments in this chapter,the pharmacological effects of Polygonum cuspidatum against oxidation and inflammation were selected as the evaluation index for the comparative study of the efficacy of Polygonum cuspidatum integrated decoction pieces and traditional decoction pieces.The efficacy of the two decoction pieces was evaluated from the perspective of in vitro antioxidant and anti-inflammatory.Through DPPH free radical scavenging experiment,ABTS free radical scavenging experiment,superoxide free radical scavenging experiment,reducing activity test and hydroxyl free radical scavenging experiment,the in vitro antioxidant activity of Polygonum cuspidatum integrated decoction pieces and traditional decoction pieces were compared.The results showed that the two kinds of decoction pieces have certain in vitro antioxidant activity,and the integrated decoction pieces of Polygonum cuspidatum have better activity.Through the in vitro model of RAW264.7 cell inflammation induced by LPS,the results showed that the alcohol extract of Polygonum cuspidatum significantly inhibited the increase in the levels of inflammatory factors IL-6,IL-1β and TNF-α induced by LPS,and the alcohol extract of decoction pieces was integrated The anti-inflammatory effect of LPS-induced RAW264.7 cell inflammation model is slightly better than that of traditional decoction pieces.4.Investigation and research on storage conditions and shelf life of Polygonum cuspidatumThe study on the storage conditions of fresh Polygonum cuspidatum and the stability of the integrated decoction pieces of Polygonum cuspidatum found that when fresh Polygonum cuspidatum is stored within 5 days,the quality loss of the medicinal materials is small,and when it reaches 15 days,the loss of 5 active ingredients and extracts is large..There was no significant change in appearance of Polygonum cuspidatum packaged in aluminum foil bag and transparent bag during the inspection period.Except for the slight increase in the content of emodin,the content of other active ingredients and extracts has decreased,and the content of active ingredients in the aluminum foil bag is less than that of the transparent bag packaging.However,the decoction pieces under the two types of packaging are under the inspection period The testing indicators are in compliance with regulations.
