| Objective: To study the effects of chemokine receptor 4(CCR4)on proliferation,invasion and migration of human clear cell carcinoma(CCRCC)cells.Methods: using Western blot(WB)and immunofluorescence technique to detect renal clear cell carcinoma cells strain 786-0,ketr-3 and kidney embryonic cell lines of 293 t cell lines CCR4 expression,CCR4 interference by transfection kidney cancer cell lines 786-0,786-0 cell lines CCR4 gene expression,stable flow cytometry screening interfere with cell lines,Western blot technical appraisal interference effect.RCC cell lines 786-0 were divided into three groups: 786-0,786-0/LV-CCR4 and 786-0/CON cells.Cell proliferation ability of the three groups was detected by MTT assay,cell migration and invasion ability were detected by cell scratch assay and Transwell invasion assay.Results: CCR4 was highly expressed in transparent cell carcinoma cell line 786-0 and ketr-3(P < 0.05).Silencing the expression of CCR4 in transparent cell carcinoma cell line 786-0 had no significant effect on its proliferation,but significantly reduced the migration and invasion ability of renal carcinoma cell line(P < 0.05).Conclusion: CCR4 is highly expressed in renal clear cell carcinoma cells.CCR4 can promote the invasion and metastasis of 786-0cell line,but has no effect on its proliferation ability.It may affect the occurrence and progression of tumors by affecting the invasion and metastasis ability of tumor cells,and CCR4 may become a potential therapeutic target of cc RCC. |
