| ObjectiveObjective to investigate the effects of naringenin on the proliferation,apoptosis and migration of tongue squamous cell carcinoma Tca8113 cells in vitro and its related pathways.MethodsMethods: Tca8113 cells were treated with naringenin at the concentrations of 0,20,40,60,80 and 100 μ mol / L for 24,48 and 72 hours,respectively.The inhibitory effect of naringenin on Tca8113 cell proliferation was detected by MTT assay,and its 50% inhibitory concentration was observed and calculated The effects of naringenin at 0,20,40,80 μ mol / L on the apoptosis of Tca8113 cells were detected by V-FITC / PI double staining;the apoptosis morphology of Tca8113 cells treated with naringenin at 0,20,40,80 μ mol / L for 24 h was observed by hoechst33342 staining;the cloning rate of Tca8113 cells treated with naringenin at 0,20,40,80 μ mol / L for 24 h was observed by plate cloning assay;Western blot analysis was used to detect the apoptosis of Tca8113 cells The expressions of p53,p21,Bcl-2,Bax,caspase9 and Caspase3 in the cells treated with naringenin at the concentrations of 0,20,40 and 80 μ mol / L for 24 h were detected by blot,and the migration ability of Tca8113 cells treated with naringenin at the concentrations of 0,20,40 and 80 μ mol / L for 24 h was detected by scratch test.ResultsMTT and annexin V-FITC / PI results showed that naringenin significantly increased the inhibition rate and apoptosis rate of Tca8113 cells in a concentration and time-dependent manner,indicating that naringenin can effectively inhibit the proliferation and promote apoptosis of Tca8113 cells,thereby inhibiting the activity of Tca8113 cells.Hoechst 33342 staining showed that with the increase of naringenin concentration,tongue squamous cell carcinoma Tca8113 cells showed different degrees of cell deep staining and nuclear shrinkage.The results of plate cloning experiment showed that naringenin could effectively reduce the clone formation ability of tongue squamous cell carcinoma Tca8113 cells in a concentration dependent manner.Western blot results showed that naringenin could enhance the expression of p53,p21,Bax,caspase9,Caspase3,and weaken the expression of Bcl-2 in a concentration dependent manner.The results of cell scratch test showed that naringenin could significantly reduce the migration rate of Tca8113 cells.ConclusionsNaringenin can effectively inhibit the proliferation of tongue squamous cell carcinoma Tca8113 cells in the range of 0-100 μ mol / L;naringenin can effectively promote its apoptosis in a concentration and time-dependent manner in the range of 0-80 μ mol / L,which may be related to the caspase cascade of p53-p21 pathway;meanwhile,naringenin can significantly reduce the apoptosis of tongue squamous cell carcinoma Tca8113 cells in the range of 0-80 μ mol / L The migration rate was in a concentration dependent manner. |
