Objective To explore the protective effect and mechanism of crocin on renal injury in diabetic rats.MethodsIn 60 male SD rats,streptozotocin(STZ)was injected intraperitoneally at a dose of 55 mg/kg to induce diabetic rats.After the model was induced successfully,diabetic rats were randomly divided into three groups: diabetic group,crocin treatment group(30 mg/kg)and metformin group(75 mg/kg,positive control).Another 10 normal rats were taken as the control group.12 weeks later,blood glucose,BUN,ACR,Scr,MDA,SOD and CAT were measured.HE staining was used to observe renal pathological changes,the relative protein expression of NF-κ B,TNF-α and IL-6 were detected by immunohistochemistryResults Compared with the control group,the levels of blood glucose,BUN,ACR,Scr,MDA,the protein expression of NF-κB,TNF-α,IL-6 were significantly increased and the content of SOD and CAT were decreased significantly in the diabetic group(P < 0.05);Compared with the diabetes group,the blood glucose,BUN,ACR,Scr,and MDA levels of the crocin treatment group and the metformin group were significantly reduced,the expression of NFκB,TNF-α and IL-6 protein was significantly decreased,and the content of SOD and CAT were significantly increased(P<0.05).Conclusion Crocin can protect the kidney of diabetic rats,and its mechanism may be related to lowering blood sugar,inhibiting oxidative stress and inflammation.
