| Objective: To investigate the relationship between interleukin-18 137 G>C,607 C>A and COX-2 1195 A>G,8473 T>C,765 G>C single nucleotide polymorphis ms(SNP)and the susceptibility of uygur adult population with chronic periodontiti s(CP)in Moyu county,Xinjiang.Methods: A total of 300 oral and buccal mucos al swab samples were collected from chronic periodontitis and healthy controls by o-ur research group’s investigation research in Moyu county during 2013(100 cases in the mild group,100 cases in the moderate to severe group,and 100 cases in t he control group).Genomic DNA was extracted and the distribution of five genoty pes of interleukin-18 and COX-2 were detected by polymerase chain reaction-restric tion fragment length polymorphism(PCR-RFLP).Results: IL-18 137 G>C site: ther e was a statistically significant in GG genotype that distributed in periodontitis gro up and mild group compared to control group(P=0.045,OR=1.67;P=0.01,OR=2.23);IL-18 137 G>C site: CC genotype distribution was statistically significant bet ween chronic periodontitis group and mild group compared to control group(P=0.045,OR=1.67;P=0.01,OR=2.23);The distribution of COX-2 1195 A>G and 8473T>C sites were only statistically significant between the moderate-severe group com pared to control group(P=0.032,OR=1.86;P=0.024,OR=0.34);And we found ther e were statistically significant distribution in 765 G>C GC genotypes both betwee n in the periodontitis group,mild group and the moderate-severe group compared t o control group(P=0.01,OR=2.12;P=0.041,OR=0.49;P=0.025,OR=2.2);Multipl e logistic regression analysis showed the age was negatively correlated with the sus ceptibility to chronic periodontitis,and age under 35 years old was a protective fac tor for chronic periodontitis(OR=0.029,95%CI=0.035-0.108,P<0.001).Logistic regr ession results showed that the C allele in 137 G>C of IL-18 and the C allele in COX-2 8473 T>C were risk factors for chronic periodontitis after matched gender and age.Conclusion: The polymorphism of IL-18 and COX-2 genes in Uygur adul ts in Moyu County,Xinjiang was correlated with the susceptibility to chronic perio dontitis,the C allele in IL-18 137 G>C and COX-2 8473 T>C may be independen t risk factors for chronic periodontitis. |
