ObjectiveTo investigate the effect and mechanism of TLR7 gene on the apoptosis of hippocampal neuronal induced by hypoxia,hypoglycemia and reoxygenation in rats.MethodsAccording to the TLR7 m RNA coding sequence,the interference si RNA sequence was designed and synthesized,and then transfected into rat hippocampal neurons.The experiment was divided into four groups: Control group,Model group,Model+TLR7-si RNA group and Model+NC group.TLR7 gene silencing was detected by q RT-PCR,the apoptotic rate was detected by Hoechst33258 staining and flow cytometry,and the relative expression of TLR7,NF-κB and Caspase-3 was detected by Western blot.ResultsCompared with the control group,the expression of TLR7 gene and protein increased significantly,the expression of Caspase-3,NF-κB and apoptotic rate increased significantly in Model group and Model+NC group(P< 0.05);compared with Model group,the expression of TLR7 gene and protein decreased significantly,while the expression of Caspase-3,NF-κB and apoptotic rate decreased significantly in Model+TLR7-si RNA group(P < 0.05).ConclusionTLR7 gene silencing can inhibit the apoptosis of hippocampal neurons induced by hypoxia,glucose deprivation and reoxygenation,and its mechanism may be related to the down-regulation of Caspase-3 and NF-κB expression.
