A Correlative Study Of The Effect Of MiRNA-125b On Proliferation,Apoptosis,Migration And Invasion Of Tongue Squamous Cell Carcinoma Cell Line Tca8113 | | Posted on:2022-07-30 | Degree:Master | Type:Thesis | | Country:China | Candidate:L Chen | Full Text:PDF | | GTID:2504306326999689 | Subject:Oral Medicine | | Abstract/Summary: | PDF Full Text Request | | Objective: To investigate the effect of microRNA-125b(miRNA-125b)on the proliferation,apoptosis,migration and invasion of tongue squamous cell carcinoma(TSCC)cell line Tca8113.Methods: Real-time fluorescence quantitative polymerase chain reaction(RT-PCR)was used to detect the expression of miRNA-125 b in TSCC tissues(15 cases)and normal tissues adjacent to cancer.The miRNA-125 b mimics group,miRNA-125 b inhibitor group,mimics NC group,inhibitor NC group and blank cell group(control group)were constructed and transfected into tongue squamous cell carcinoma cell line Tca8113,respectively.CCK-8 assay was used to detect cell proliferation,scratch test was used to detect cell migration ability,Transwell tset was used to detect cell invasion ability,and flow cytometry was used to detect cell apoptosis.Results: The average expression level of miRNA-125 b in TSCC tissues was significantly lower than that in adjacent normal tissues(P < 0.001).Compared with the control group,there was no significant difference in cell proliferation,apoptosis,migration and invasion ability between mimics NC group and inhibitor NC group(P>0.05).Compared with the control group,the miRNA-125 b mimics group had significantly reduced cell proliferation,migration and invasion ability,and the apoptosis rate increased significantly;while the miRNA-125 b inhibitor group cell proliferation,migration and invasion ability increased significantly,cell apoptosis rate significantly decreased(P<0.001).Conclusion: The average expression level of miRNA-125 b in TSCC is significantly lower than that in adjacent tissues,and the high expression of miRNA-125 b in Tca8113 cells can inhibit its proliferation,migration and invasion ability and promote its apoptosis,so miRNA-125 b may be a treatment potential targets of TSCC. | | Keywords/Search Tags: | miRNA-125b, TSCC, proliferation, apoptosis, migration and invasion | PDF Full Text Request | Related items |
