Study On The Protective Mechanism Of Yuye Decoction On Kidney Of T2DM Rats Through AGEs/RAGE

Objective: By detecting the effects of blood glucose,blood lipids,glycosylated hemoglobin,renal pathological changes and AGEs/RAGE expression in T2 DM rats,reveal the mechanism of Yuye Decoction on kidney protection in T2 DM rats and the correlation of AGEs/RAGE signaling pathway.Methods: 100 healthy male SD rats aged 3 weeks were selected for the experiment,fed with high-fat and high-sugar feed combined with low-dose STZ to establish a T2 DM rat model.The adult rats were randomly divided into model group,Yuye Decoction high-dose group,Yuye Decoction low-dose group and metformin group.Ten rats of the same batch of the same age were reserved as normal control group.Animals in each group were reared in separate cages with normal water intake.Except for the normal group,all animals were continuously fed with high-fat and high-sugar feed for 8 weeks.Gavage treatment at a fixed time every day,and regularly check body weight and blood sugar every week.After 8 weeks of treatment,the rats were sacrificed and related indicators were detected.HE staining was used to observe the pathological changes of rat kidney tissue,immunohistochemistry was used to detect the expression of RAGE protein in rat kidney tissue,ELISA was used to detect the content of AGEs in rat kidney tissue,and Western blot was used to verify the expression of RAGE in kidney tissue.Results:1.Compared with the model group,the weight,blood glucose,blood lipids,and glycosylated hemoglobin of rats in each treatment group were significantly improved (P<0.05);2.Compared with the normal group,the renal corpuscles of the model group increased in size,the number of cells in the glomerulus increased,the renal tubular epithelial cells were edema,the cytoplasm was loosened,balloon-like changes appeared,and inflammatory cell infiltration appeared in the interstitium.Part of the renal tubules is necrotic,nucleus lysis occurs in the renal tubular epithelial cells,and the cytoplasm turns red.After the intervention of Yuye Decoction,the renal tissue morphology of rats in each treatment group was significantly improved;3.Compared with the model group,the expression of AGEs and RAGE protein in the kidney tissue of rats in each treatment group was significantly reduced(P<0.05);ased(P < 0.05);Conclusion: Yuye Decoction can effectively reduce blood sugar,blood lipids,glycosylated hemoglobin,and reduce the expression of AGEs and RAGE in the kidney tissue of T2 DM rats.Yuye Decoction has certain preventive and therapeutic effects on the kidneys of T2 DM rats,and its mechanism of action may be related to the inhibition of the AGEs-RAGE signaling pathway in the kidney tissue.
