| BackgroundPaget’s disease(PD)is a rare malignant skin tumor,often manifested by pruritus,erythema,erosion and eczema-like symptoms,so it is also called eczema-like cancer.According to the location of the disease,it can be divided into two types:Mammary Paget’s disease(MPD)with skin lesions in the nipple and areola;Extramammary Paget’s disease(EMPD)is known as extramammary Paget’s disease.P53,the tumor suppressor protein most associated with humans,is mainly responsible for inducing cell apoptosis and cell growth arrest.The apoptosisstimulating protein family of P53,which contains ASPP1(apoptosis stimulating proteins of P531),ASPP2(apoptosis stimulating proteins of P53 2),and IASPP(inhibitory member of the ASPP family),can specifically bind to P53 proteins to promote or prevent their pro-apoptosis.Therefore,the abnormal expression of ASPP1,ASPP2,and iASPP in the ASPP family may be related to the formation and development of tumors.However,there are no reports about the expression of ASPP family in EMPD and MPD tissue home and abroad.ObjectiveImmunohistochemical SP method was used to detect the expression of ASPP family members ASPP1,ASPP2,iASPP in EMPD and MPD tissue and the mechanism of action of these three factors in the pathogenesis of EMPD and MPD and its clinical significance were analyzed.MethodsWe use immunohistochemical to detect the expressions of ASPP1,ASPP2 and iASPP in 20 EMPD tissues,20 MPD tissues and 10 normal skin tissues.SPSS 25.0was used for analysis,in which ANOVA was used to describe the expression of ASPP1,ASPP2 and iASPP in EMPD,MPD and normal skin tissues,and Pearson was used to analyze the correlation of the three factors in different tissues.A =0.05 was the test level,and P<0.05 indicated statistically significant.Results1.The expression of ASPP1 in these three tissues: EMPD tissue(4.104±0.948),MPD tissue(4.265±0.673),normal skin tissue(3.818±0.700).There was no significant difference between the three groups(F=1.041,P>0.05).2.The expression of ASPP2 in these three tissues: EMPD tissues(3.095±1.079),MPD tissues(3.397±0.728),and normal skin tissues(3.318±0.376).There was no significant difference between the three groups(F=0.668,P>0.05).3.The expression of IASPP in these three tissues: EMPD tissues(6.340±0.594),MPD tissues(8.701±0.699),normal skin tissues(3.296±0.532).The difference between the three groups was statistically significant(F=226.473,P<0.001):the expression of IASPP in EMPD was significantly lower than MPD and significantly higher than that in normal skin tissue(all P< 0.01).4.Pearson correlation analysis: there was no correlation between the expression of ASPP1,ASPP2 and IASPP in EMPD,MPD and normal skin tissues(P> 0.05).Conclusions1.The expression of ASPP1 and ASPP2 was low in EMPD,MPD and normal skin tissue,and there was no significant difference in the expression of ASPP1 and ASPP2 in the three tissues,suggesting that ASPP1 and ASPP2 may not participate in the occurrence of EMPD and MPD;2.IASPP was highly expressed in EMPD and MPD,and was higher in MPD than EMPD,suggesting that IASPP may participate in and promote the occurrence and development of EMPD and MPD. |
