Liver cancer,also known as liver malignant tumor.The incidence rate of liver cancer is the fifth place in China.It ranks sixth in the world.It is a global health challenge.Its etiology and exact molecular mechanism are not fully understood.Primary liver cancer can be divided into primary and secondary types.Primary liver cancer originates from the epithelial or mesenchymal tissue of the liver and is one of the most common and fatal malignant tumors in the world.Hepatocellular carcinoma is a kind of primary liver cancer with high mortality,accounting for about 90% of primary liver cancer.It is the third most common cause of cancer-related mortality in the world.However,in the process of the formation of hepatocellular carcinoma,it is usually from chronic hepatitis to cirrhosis,early hepatocellular carcinoma,and finally to advanced hepatocellular carcinoma.In this paper,we analyzed the potential relationship between differential gene expression analysis and weighted gene co-expression network analysis(WGCNA).The former is mainly to find the differential expression changes of a single gene in the transformation process,while the latter is to find the key modules and their genes related to the transformation process from the perspective of module genes.In this paper,we used GSE114564,a hepatocellular carcinoma related gene expression data set in GEO database.The data set included 15 normal liver samples,20 chronic hepatitis samples,10 liver cirrhosis samples,18 early hepatocellular carcinoma samples and 45 advanced hepatocellular carcinoma samples,a total of 108 samples.We first analyzed the differentially expressed genes,and got 130 differentially expressed genes between chronic hepatitis and normal liver,212 differentially expressed genes between cirrhosis and normal liver,241 differentially expressed genes between early hepatocellular carcinoma and normal liver,and 784 differentially expressed genes between advanced hepatocellular carcinoma and normal liver.Secondly,weighted gene co-expression network analysis was used to construct the co expression network.30 modules were obtained,and the key modules were found according to the gene expression of different traits.The relationship between key modules and different stages of HCC was identified,and the most significant modules were selected for analysis.Then,we match the differential gene with the key module gene to find the common gene.Finally,the metabolic pathways in different stages of HCC transformation were analyzed.
