Purpose: Through a preliminary search on CNKI,it was found that both donkey-hide glue and deerhorn glue can play a role in enriching blood in the human body.Using donkey-hide glue and deerhorn glue as raw materials,after digestion with trypsin,the donkey-hide gelatin polypeptide and staghorn gum polypeptide were obtained by ultrafiltration.In this experiment,the activity of donkey-hide gelatin polypeptide and staghorn gum polypeptide on aplastic anemia mice was evaluated.deerhorn glue is a substitute for donkey-hide glue in the treatment of aplastic anemia,providing reference and experimental basis for the treatment of aplastic anemia in Chinese medicine in the future.Method:According to the identification items and content determination items of donkey-hide gelatin and deerhorn glue in the Pharmacopoeia of the People’s Republic of China in 2020,using L-hydroxyproline as a reference,one measurement and multiple evaluation methods,realize the determination of L-proline,glycine and alanine content,achieve the purpose of quality analysis of the purchased donkey-hide glue and deerhorn glue;Take the degree of hydrolysis as the inspection index,and use the method of single factor analysis to investigate the enzymatic hydrolysis process of donkey-hide glue and deerhorn glue;With the help of ultrafiltration separation method,the separation of peptides(molecular weight less than or equal to 10 KDa)after enzymatic hydrolysis of donkey-hide gelatin and deerhorn glue can be achieved;By comparing the pharmacodynamic activity of donkey-hide gelatin polypeptide and staghorn gum polypeptide on cyclophosphamide(CTX)-induced aplastic anemia model mice,the number of red blood cells(RBC)and hemoglobin(Hb)of the model mice were compared.The number is used as an indicator to investigate the substitution of deerhorn glue for donkey-hide glue in the treatment of aplastic anemia model mice.Result:Refer to the identification items and content determination items in the 2020 edition of "The Pharmacopoeia of the People’s Republic of China",the study found:There is no obvious difference between donkey-hide glue and deerhorn glue in properties,there are differences in color and smell,donkey-hide glue is brown or dark brown,with a slight smell and sweetness,deerhorn gum is yellowish brown or reddish brown,slightly sweet and salty in smell;the content of 4 free amino acids in donkey-hide glue and deerhorn glue was determined by the one-test-multiple-assessment method,the study found:All the drugs purchased in the experiment meet the content requirements stipulated in the2020 edition of the Pharmacopoeia of the People’s Republic of China.The research on the enzymatic hydrolysis process of donkey-hide glue and deerhorn glue found:First,the donkey-hide glue or deerhorn glue and water are turned at a ratio of 1:50 at80 ℃.Then use 1% sodium hydroxide solution to adjust the solution to p H=8,add 5%trypsin(the proportion of trypsin to the weight of the original drug),when the enzymatic hydrolysis temperature is adjusted to 37 ℃ and the enzymatic hydrolysis time is 4 h,then it was cooled to room temperature and centrifuged in a high-speed refrigerated centrifuge at 4500 r/min and 4 ℃ for 10 min,take the supernatant,the enzymatic hydrolysate of donkey-hide glue and deerhorn glue,Finally,boil on a water bath for 5 min to terminate the enzymatic hydrolysis.The research on the separation of donkey-hide gelatin polypeptide and staghorn gum polypeptide found:Transfer the enzymatic hydrolysate of donkey-hide glue and deerhorn glue to an ultrafiltration tube with a molecular weight cut-off of 3 KDa,then place it on a high-speed refrigerated centrifuge at 4000 r/min and centrifuge at 4 ℃ for 5 min,That is,donkey-hide gelatin polypeptide and staghorn gum polypeptide with molecular weights below 10 KDa are obtained.A mouse model of aplastic anemia induced by cyclophosphamide was established,and the pharmacodynamics of A collagen solution,donkey-hide gelatin polypeptide solution,staghorn collagen solution and staghorn gum peptide solution were studied and found:Compared with the blank control group,the A collagen solution group,the donkey-hide gelatin peptide group,the staghorn collagen solution group and the staghorn glue peptide group all have good blood-enriching effects;Compared with the donkey-hide gelatin peptide group,the Lujiaojiao peptide group P<0.05.Conclusion:Through the comparative study of the pharmacodynamics of donkey-hide glue and deerhorn glue against aplastic anemia in mice,it was found that the deerhorn glue has the potential to replace donkey-hide glue.
