Objective In this experiment,the poorly water-soluble total triterpenes from Poria cocos cortex was prepared into liposome preparations,in order to improve the shortcomings of poor water solubility,and modified with hyaluronic acid to improve the targeting and stability of liposomes,and stablished a preliminary quality evaluation system for its freeze-dried preparations to provide reference for follow-up treatment of urinary system diseases.Methods A method for content determination by HPLC was established,and the method for measuring encapsulation efficiency and preparation method of liposomes were screened.Moreover,the formulation of liposomes were preferred by single factor and orthogonal experiment;The single factor method was used to optimize the preparation process of HA modified Poria cocos cortex total triterpene liposomes and determine the freeze-drying process of HA modified Poria cocos cortex total triterpene liposomes;We not only evaluated the quality of liposome freeze-dried preparations from the appearance,particle size,Zeta potential,PDI,drug loading,encapsulation rate,content,but also investigated the leakage rate and invitro release of liposome.Results The optimal formulation of hyaluronic acid-modified Poria cocos total triterpene freeze-dried liposomes is : the mass ratio of soybean lecithin-cholesterol is 7:1,the drug-lipid ratio is 1:3,the p H of the PBS is 7.0,the concentration of HA is 0.5mg/m L,and the magnetic stirring speed is 650r/min,the ultrasound time of the probe cortex is 2 min,the mass ratio of mannitol to liposomes is 1.5:1,the lyophilized liposome prepared by the prescription is soft and plump with white powder and the appearance after reconstitution is light with white transparent clear liquid.And the particle size,Zeta potential,PDI,drug loading,encapsulation efficiency,content and other aspects of liposome meet the expected requirements.They are suitable for storage at 4 degrees and more stable than liposomes without HA modification.Conclusions We established the method for determining the content and encapsulation efficiency of total triterpene liposomes from Poria cocos cotex;The preparation process of total triterpene liposomes modified by hyaluronic acid from Poria cocos was confirmed,with good reproducibility;The quality standard for the freeze-dried liposomes of total triterpene from Poria cocos was initially established and achieved the purpose of the experiment.
