Objective To explore the relationship between the expression level of tripartite motif 59(TRIM59)in hepatocellular carcinoma(HCC)cells and the classic molecule E,N-cadherin in epithelial-mesenchymal transition(EMT)in tumor invasion and migration.Methods Six HCC cell lines(Huh7,Hep3 B,Hep G2,SK-Hep1,BEL7402,SMMC7721)and normal human liver cell line LO2 were purchased as experimental research objects.Real-time fluorescent quantitative PCR(qRT-PCR)was used to detect the expression of TRIM59 mRNA in each HCC and LO2 cell lines,and the protein expression of TRIM59 in the experimental cells was measured by Western Blot.The HCC cell line with relatively high expression of TRIM59 mRNA and protein in the experiment was selected as the cell line required for the later experiment.A cell line with down-regulated expression of TRIM59 was co-cultured with anti-TRIM59,and a cell line with up-regulated expression of TRIM59 was cultivated by infection with a lentivirus carrying the TRIM59 plasmid(pc DNA3.1-TRIM59).Western blot experiments were performed to detect the expression of N-cadherin and E-cadherin before and after TRIM59 interference.Scratch test and Transwell test detect the invasion and migration ability of HCC before and after TRIM59 interference.Results The results of qRT-PCR and Western Blot analysis showed that the mRNA and protein expression of TRIM59 in HCC cell lines Huh7,Hep3 B,Hep G2,SK-Hep1,BEL7402,SMMC7721 was significantly higher than that of normal human liver cell line LO2(P All <0.05).In the cell line where TRIM59 protein was interfered,the expression of E-cadherin decreased and the expression of N-cadherin increased;in the cell line where TRIM59 protein was overexpressed,the expression of E-cadherin increased,and the expression of N-cadherin increased Decrease(all P<0.05).Scratch test and Transwell test showed that high expression of TRIM59 can significantly enhance the invasion and migration ability of hepatocellular carcinoma(P<0.05).Conclusions TRIM59 mRNA and protein are highly expressed in hepatocellular carcinoma,which may increase the expression of E-cadherin and decrease the expression of N-cadherin,the classical molecule in EMT,to promote the invasion and migration of hepatocellular carcinoma.TRIM59 is very likely to become another new target for precision treatment of liver cancer.
