| Background and Purpose:Hypoxia is an important factor involved in regulating gene expression,especially the hypoxia-inducible factor-1(HIF-1)gene,which is an important regulator of oxygen homeostasis in almost all mammals.Hypoxia is an important factor that changes the intestinal environment.The body triggers the adaptation to hypoxia by activating HIF.HIF-1 is widely expressed in the microenvironment of tumor cells and plays a key role as a regulatory subunit of oxygen homeostasis in many cell types.GLUT-1 and CA9 are downstream genes regulated by HIF-1.GLUT-1 is involved in regulating the transport of glucose on the cell membrane.CA9 mainly maintains the body’s acid-base balance.Since the growth of the tumor precedes the development of its blood vessels,a hypoxic zone is often formed in the tumor itself.These areas can occur either at the edge of the tumor with their spreading ability,or at the center of the tumor due to insufficient blood vessel penetration.This study mainly explores the expression of HIF-1,GLUT-1,CA9 in colorectal cancer and its relationship with the clinical characteristics of tumor patients,and provides a reference for the diagnosis and treatment of colorectal cancer.Methods:The samples were derived from 64 cases of paraffin-embedded specimens diagnosed as colorectal cancer in the Department of Gastrointestinal Surgery,China-Japan Union Hospital of Jilin University from January 2014 to December 2015.The tumor and its adjacent tissues(from the edge of the cancerous tissue)Above 5cm).Use adjacent tissues as controls to observe the expression of HIF-1,GLUT-1,and CA9 in tumor tissues.Statistics of the clinical data of 64 patients,including: gender,age,maximum tumor diameter,location,stage,degree of differentiation,depth of invasion,lymph node metastasis,nerve vascular invasion,overall survival and other11 indicators,analyzed three indicators The relationship between markers and clinicopathological features.Results:The expression rates of HIF-1,GLUT-1,and CA9 among 64 patients were79.7%,87.5%,and 84.3%,respectively;HIF-1 and tumor differentiation were statistically different,and GLUT-1 and tumor infiltration depth were different There are statistical differences,CA9 and tumor location are statistically different;HIF-1,GLUT-1,and CA9 are all correlated and positively correlated;HIF-1 is correlated with 5-year overall survival Statistical difference.Conclusions:1.HIF-1,GLUT-1 and CA9 all have a high degree of positive expression in 64 colorectal cancer tumor tissues,which can be used as one of the indicators of tumor diagnosis.2.The expression of HIF-1,GLUT-1,and CA9 has a certain correlation with the clinicopathological characteristics of colorectal cancer,which can guide the prognosis and treatment of tumors.3.HIF-1,GLUT-1 and CA9 are related,and it can be inferred that they have a synergistic effect in the occurrence and development of tumors.4.Colorectal cancer patients with moderate/high HIF-1 expression have poor prognostic survival. |
