| Objective: Screen drugs with good repellent effects on leeches,adopt different dosage forms of rapid release and sustained release,and prepare new repellent formulations with different drug release rates,aiming to realize rapid-acting,sustained-release and other multi-phase drug release modes,cover all the repellent stages of leeches and prolong the repelling time of drugs.Methods: The repellent,contact and fumigation effects of various essential oils on Hirudo nipponica Whitman were investigated,and the most effective essential oils were selected from various aspects.Gelatin and sodium alginate were used as capsule materials by complex coacervation method to prepare tea tree oil microcapsules,Konjac glucomannan and ethyl cellulose were used to prepare tea tree oil emulsion,and glycerylmonooleate was used to prepare tea tree oil lyotropic liquid crystals.And their characterization were performed separately.Filter paper circle method and water repellent experiment were committed to evaluate the repellent effects of different preparations on Hirudo nipponica Whitman.Measure the activities of acetylcholinesterase,glutathione S-transferase and carboxylesterase enzymes in the leech’s body body,aiming to explore the mechanism of tea tree oil on Hirudo nipponica Whitman from the biochemical level.Results: In the study,Hirudo nipponica Whitman was used as the test animal.Firstly,a variety of plant essential oils were screened,and tea tree oil with good repellent effect on Hirudo nipponica Whitman was selected.Because tea tree oil is volatile and unstable in physical and chemical properties,tea tree oil was prepared into microcapsules,emulsion and lyotropic liquid crystals to protect it in the study.A series of characteristics of various preparations were investigated.Finally,it was proved that tea tree oil emulsion and tea tree oil microcapsules could effectively repel Hirudo nipponica Whitman and prolong the time of tea tree oil repellent by the behavioral experiments and molecular biological evaluations.But tea tree oil lyotropic liquid crystals had almost no effect on Hirudo nipponica Whitman repelling.Conclusion: Tea tree oil has a good repellent effect on Hirudo nipponica Whitman,and tea tree oil emulsion and tea tree oil microcapsules have the same good repellent effect,which can prolong the repelling time of tea tree oil.In summary,this study provides safe,effective and easy-to-use pharmaceutical preparations to provide new options for the repellent of leeches. |
