| Objective: To investigate the expression of FOXO1 in chronic periapical periodontitis by collecting periapical samples from patients with chronic periapical periodontitis,and to provide reference for exploring the role and mechanism of FOXO1 in chronic periapical periodontitis.Method: The tissues collected were divided into experimental group(15 cases)and control group(15 cases)according to granulation tissue of chronic periapical periodontitis and healthy periapical periodontitis,and immunohistochemical staining and real-time PCR were performed.1.Clinical sample collection The experimental samples were collected from clinical patients admitted to the Stomatological Hospital Affiliated to School of Stomatology of Dalian Medical University between July 2019 and July 2020.The experimental group was clinically diagnosed with chronic periapical periodontitis and needed apical surgery or tooth extraction.The control group was the healthy premolars that needed to be removed for orthodontics or the third molars that needed to be removed because of the impacted third molars.All samples were collected with informed consent of the patients and approved by Dalian Medical University ethics committee.2.FoxO1 protein expression was detected by immunohistochemical staining Five samples from the experimental group and control group were fixed,dehydrated,and paraffin sections were prepared by dipping wax after tissue transparency.The slices are dried,dewaxed,dyed and sealed.The average optical density of FoxO1 was measured and calculated.SPSS Statistics22.0 software was used for statistical analysis of the results.3.Real-time PCR was used to detect the expression of FoxO1 gene The 10 samples of the experimental group and the control group were respectively ground and extracted RNA,and the RNA concentration was measured.The RNA was reverse transcribed into c DNA,and the expression of FoxO1 m RNA was detected by SYBR Green method.Collecting initial data,analysis of determination of FOXO1 and GAPDH Ct value,using comparing 2-ΔΔCt cycle threshold value method,to analyze and determine the relative amounts FOXO1 gene expression in each group.Results: 1.Collection of clinical samples The experimental group included 8 males and 7 females,aged 24 to 70 years.There were 4 males and 11 females in the 15 control group,aged 19 to 72 years.2.Immunohistochemical staining results Immunohistochemical staining showed that FoxO1 was expressed in periapical tissue samples of both groups,and the expression of FoxO1 protein in the experimental group was higher than that in the control group,and the difference between the two groups was statistically significant(P < 0.05).3.Real-time PCR results FoxO1 was expressed in both the experimental group and the control group,and the FoxO1 gene expression in the experimental group was higher than that in the control group,and the difference between the two groups was statistically significant(P<0.05).Conclusion: FoxO1 expression is increased in periapical tissues of patients with chronic periapical disease,suggesting that FoxO1 is involved in the disease process of chronic periapical disease. |
