| Objective: To investigate the differential expression of LncRNA PTPRD-AS1 in ovarian cancer and its effect on cell biological behavior and its mechanism.Methods: 1.Database and bioinformatics analysis1.1 The target gene LncRNA PTPRD-AS1 was understood through online database,and the upstream regulatory gene of LncRNA PTPRD-AS1 was searched.1.2 Er RNA-Seq expression profiles of ovarian cancer and normal ovarian tissue were downloaded from cancer TCGA and GTEX databases,respectively.Bioinformatics methods were used to integrate the data sets,select the target genes and analyze their differential expression in ovarian cancer and normal tissue.1.3 The prognostic value of the target gene was determined by Kaplan-Meier survival analysis and Cox regression analysis,and the KEGG pathway was analyzed by GSEA.2.Experimental verification2.1 Clinical specimen collectionTumor tissue and corresponding normal ovarian tissue were collected from 15 gynecological patients with high-grade serous ovarian cancer in The First Affiliated Hospital of Wannan Medical College from September 2018 to September 2019.Tissues were immediately stored in liquid nitrogen for refrigerated transportation and stored at-80 ℃.None of the patients enrolled in this study had received chemotherapy or radiotherapy.Our study was approved by the hospital ethics committee and received informed consent from all patients.2.2 RT-q PCR was used to verify the expression level of lnc RNA PTPRD-AS1 in 15 ovarian cancer tissues and normal ovarian tissue samples,ovarian cancer cell line H08910 and human normal ovarian cell line HOEPIC as control.2.3 After up-regulating or down-regulating the expression of lnc RNA PTPRD-AS1 in H08910 cells by cell transfection,the invasion,migration and apoptotic functions of cells were detected and analyzed by scratch healing method,Transwell method and apoptosis staining,respectively.Results: 1.Database and bioinformatics analysis results1.1 The target gene lnc RNA PTPRD-AS1 is the antisense RNA of the protein-coding gene PTPRD.The upstream gene of lnc RNA PTPRD-AS1 is PTPRD.1.2 The data of 379 ovarian cancer samples and 88 normal ovarian tissue samples were obtained from TCGA database and GTEX database,respectively.1.3 LncRNA PTPRD-AS1 was low expressed in ovarian cancer(P<0.05).However,the 5-year overall survival rate analysis showed that the difference between the high expression and low expression of lnc RNA PTPRD-AS1 was not statistically significant(P > 0.05).1.4 Univariate Cox regression showed that age,stage and lnc RNA PTPRD-AS1 were independent prognostic markers,and the HR of lnc RNA PTPRD-AS1 was1.591(95%CI: 1.10--2.285,P<0.05).After controlling for clinical characteristics,lnc RNA PTPRD-AS1 was still an independent prognostic marker(HR=1.532,95%CI=1.05-2.232,P<0.05).1.5 KEGG-GSEA pathway analysis showed that lnc RNA PTPRD-AS1 was involved in a series of metabolic pathways,including aminoacyl-t RNA biosynthesis,Glyoxylate and dicarboxylate metabolism,etc.LncRNA PTPRD-AS1 is associated with Bladder cancer,Colorectal cancer,and Pancreatic cancer.LncRNA PTPRD-AS1 is involved in the classical tumor-related pathways,such as the MAPK signsling pathway,the Wnt signaling pathway(a large family of secreted glycoproteins)and the transforming growth factor-β(TGF-BETA)pathway,in which the Wnt signaling pathway and TGF-BETA are the classical pathways associated with the development of ovarian cancer(P<0.05).2.Experimental verification results2.1 Compared with normal ovarian tissues and normal ovarian cells,the expression of lnc RNA PTPRD-AS1 was significantly different in ovarian cancer tissues and cells(P<0.05),and the expression was low in all of them.2.2 Upregulation of lnc RNA PTPRD-AS1 can significantly inhibit the invasion and migration of HO8910 cells,and promote the apoptosis of ovarian cancer cells.Conclusion: 1.Database and bioinformatics analysis showed that lnc RNA PTPRD-AS1 was low expressed in ovarian tissue,but 5-year overall survival analysis showed that the difference between the high expression and low expression of lnc RNA PTPRD-AS1 was not statistically significant.(P > 0.05)2 Database and bioinformatics analysis showed that the functions of lnc RNA PTPRD-AS1 were mainly involved in multiple metabolic pathways,as well as related to multiple pathways involved in the occurrence and development of ovarian cancer.3 Database and bioinformatics analysis showed that lnc RNA PTPRD-AS1 could be an independent prognostic factor for ovarian cancer.4 Low expression of lnc RNA PTPRD-AS1 in ovarian cancer.Upregulation of lnc RNA PTPRD-AS1 can inhibit the invasion and migration of ovarian cancer cells,and promote cell apoptosis.5 LncRNA PTPRD-AS1 may become a new target for the treatment of ovarian cancer,which has certain research significance. |
