Study On The Effect Of Ziyin Shugan Decoction On Hypothalamus-Pituitary-Gonadal Axis In Female Precocious Puberty Rats Based On Tiangui Theory

Objective To observe whether Ziyin Shugan Decoction treats CPP by inhibiting the premature activation of the Hypothalamus-pituitary-gonadal axis(HPGA).Methods This experiment established a female CPP rat model,intervening by low and high dose Ziyin Shugan Decoction,which to observe the vaginal opening and the histological changes of the uterus and ovary,to detectthe serum Luteinizing hormone,(LH)and Follicle stimulating hormone(FSH)levels by ELISA,and to detect the hypothalamus gonadotropin-releasing hormone(GnRH)mRNA and pituitary gonadotropin-releasing hormone receptor(GnRH-R)mRNA expression by qPCR.All indicators comprehensively reflect to observe the effect of Ziyin Shugan Decoction on the HPGA of female precocious puberty.Results Compared with the normal group,the expression of GnRH mRNA in the hypothalamus in the model group was significantly increased(P<0.01),pituitary GnRH-R mRNA expression increased,but there was no statistical difference(P>0.05);compared with the model group,the uterine index and ovarian index of the low-dose and high-dose groups of ZYSG were significantly reduced(P<0.01),LH and FSH levels were significantly reduced(P<0.05,P<0.01),hypothalamus GnRH mRNA expression in the low-dose group of ZYSG was significantly reduced(P<0.01),hypothalamus GnRH mRNA expression and pituitary GnRH-R mRNA expression in high-dose group of ZYSG was significantly decreased(P<0.01)。Conclusion Under the guidance of Tiangui theory,Ziyin Shugan Decoction has a therapeutic effect on female CPP rats,which can delay the development of sexual organs,reduce the secretion of LH and FSH,and reduce hypothalamus GnRH mRNA expression and pituitary GnRH-R mRNA expression.The effective mechanism of it may be mainly achieved by inhibiting the activation of HPGA and delaying the onset of puberty in precocious puberty rats.
