Objective: To explore the changes in the contents of the four energy substances of protein,fat,glycogen and glucose in the body of Oncomelania hupensis under laboratory conditions and temperature stress(high temperature and low temperature),as well as the two main enzymes related to aerobic respiration-amber Changes in the content of succinate dehydrogenase(SDH)and cytochrome oxidase(CCO),then to explore the effects of temperature stress on the changes in energy consumption and aerobic metabolism levels in the body of Oncomelania hupensis during the temperature stress process,to further improve the basic data on the physiological and biochemical changes of the snails under environmental stress(under low temperature or high temperature),for continuous in-depth research.Learn about Oncomelania and provide reference for further control of Oncomelania.Methods: 1.Oncomelania hupensis were collected on the branch bank of Qingyi River in Wanzhi District,Wuhu City.Firstly,the snails were washed repeatedly with tap water to remove the soil,and then divided into three groups randomly: high temperature group,low temperature group and control group.And all of them were placed in petri dishes for 3 days.Each group was divided into three parallel groups,which were numbered as group 1,group 2 and group 3.The low temperature group and the high temperature group were put into the shaker respectively,and the high temperature or low temperature stress state was achieved by controlling the temperature.The indoor feeding method of the control group was slightly improved according to the coarse straw paper feeding method as follows: Oncomelania hupensis were placed in an enamel tray of a certain specification,the bottom of the tray was padded with a layer of sponge,and then a layer of coarse straw paper was added.Natural river water was added during the feeding process to keep the coarse straw paper at a certain humidity,and the coarse straw paper was placed at room temperature of 20℃ for domestication,feeding and reserve.2.The Oncomelania hupensis of each group was quickly put into the refrigerator at-80℃freezing storage.Then take out the Oncomelania hupensis,dissect the snails with forceps crisscross method after thawing,and quickly dissect the snails under a microscope,remove the snail shell,then take out the soft tissue and place it in the pre-prepared EP tube,and the values were grouped,weighed and recorded.According to the experimental requirements of each group,add physiological saline,physiological saline or PBS buffer,etc.,homogenize the tissues,and put labels on each group and put them in the refrigerator at-80℃ for later use.3.The contents of protein,glucose,triglyceride,total cholesterol,glycogen and the activity of CCO and SDH enzymes were determined according to the kit instructions.The measurement was repeated for 3 times in each group and the arithmetic mean value was taken.Result: 1.Energy material content in O.hupensis(1)The protein contents in the soft tissue of O.hupensis were(82.36±7.92)μg/mg,(58.42±0.74)μg/mg and(63.10±0.90)μg/mg at the control,low temperature and high temperature group,respectively.The contents of protein in the three groups were statistically significant(F = 22.341,P < 0.05).(2)The glycogen contents in the soft tissue of O.hupensis were(14.20±0.42)μg/mg,(11.99±0.65)μg/mg and(12.12±0.35)μg/mg at the control,low temperature and high temperature group,respectively.The contents of glycogen in the three groups were statistically significant(F = 19.027,P < 0.05).(3)The glucose contents in the soft tissue of O.hupensis were(2.47±0.06)μg/mg,(2.45±0.07)μg/mg,(2.53±0.06)μg/mg at the control,low temperature and high temperature group,respectively.There were no statistically significant in glucose content among the three groups(F = 2.224,P > 0.05).(4)The total cholesterol contents in the soft tissue of O.hupensis were(1.39±0.05)μg/mg,(1.16±0.09)μg/mg,and(1.24±0.09)μg/mg at the control,low temperature and high temperature group,respectively.The contents of the total cholesterol in the three groups were statistically significant(F = 7.247,P < 0.05).(5)The triglyceride contents in the soft tissue of O.hupensis were(5.29±0.45)μg/mg,(3.90±0.37)μg/mg and(7.35±0.55)μg/mg at the control,low temperature and high temperature group,respectively.There was statistically significant among the three groups of triglyceride contents(F = 42.484,P < 0.05).2.Study on the influence of the contents of two main respiratory enzymes in Oncomelania hupensis(1)The contents of cytochrome oxidase in the soft tissue of O.hupensis were(27.11±2.31)μg/mg,(21.90±0.32)μg/mg,and(21.42±1.84)μg/mg at the control,low temperature and high temperature group,respectively.There was statistically significant in CCO contents among the three groups(F =10.145,P < 0.05).(2)The contents of succinic dehydrogenase in the soft tissue of O.hupensis were(1.30±0.002)μg/mg,(1.24±0.05)μg/mg and(1.12±0.02)μg/mg at the control,low temperature and high temperature group,respectively.There was statistically significant in SDH contents among the three groups(F =24.624,P < 0.05).Conclusion:(1)Under temperature stress,the contents of total protein,glycogen and total cholesterol in the soft tissue of Oncomelania hupensis were decreased.The content of triglyceride in the soft tissue of Oncomelania hupensis were variable,the content were the lowest under low temperature while the high temperature increased.This change was different from changes in other energy materials.The glucose contents in the soft tissue of Oncomelania hupensis kept relatively constant under temperature stress.(2)Temperature stress state of SDH and CCO contents in the soft tissue of Oncomelania hupensis had changed,both high temperature stress and low temperature stress all decreased the contents of CCO and SDH.
