Objective: To study the relationship between TGF-β1mRNA expression and Smads protein expression in renal tissues of rats after 5/6nephrectomy and renal fibrosis,use the medicine intervention of Jianpi Yishen Decoction to explore the protective effect of Jianpi Yishen Decoction on the kidneys and Possible regulatory mechanism.Methods: Male SD rats were randomly divided into sham operation group,renal failure model group and Jianpi Yishen Recipe treatment group.Except for the sham operation group,the other 2 groups underwent 5/6 nephrectomy to establish renal failure model.One weeks after the model was successfully established,the three groups were given the same amount of normal saline or Jianpi Yishen Recipe Concentrate(Crude Medicine 2.81g/d)by gavage every day,and the renal function,urine protein quantification and serum cells of each group were observed.The outer matrix is the main components of hyaluronic acid,laminin,type Ⅲ procollagen,type Ⅳ collagen content and TGF-β1 mRNA and Smad2,Smad3 and Smad7 protein expression levels;the pathological sections of kidney tissue were stained with HE to observe the fibrosis of the kidney tissue degree.Results: The kidneys of the three groups of rats all had the expression of TGF-β1 mRNA and Smad2,Smad3 and Smad7 protein.The expression of TGF-β1 mRNA and Smad2,Smad3 protein was weak in the sham operation group,and the expression of Smad7 protein was strong.Compared with the sham operation group,the TGF-β1 mRNA and Smad2 and Smad3 protein expressions in the renal failure model group and the Jianpi Yishen Recipe treatment group were up-regulated,and the Smad7 protein expression was down-regulated(P<0.05),the difference was statistically significant;Compared with the renal failure model group,the TGF-β1 mRNA and Smad2 and Smad3 protein expression in the Jianpi Yishen Decoction treatment group were weak,and the Smad7 protein expression was strong(P<0.05),and the difference was statistically significant.Conclusion: Jianpi Yishen Recipe delay the process of renal fibrosis by inhibiting the TGF-β1/Smad signal transduction pathway in rat kidney tissue.
