Objective:In vitro experiments were conducted to observe the effect of Guben Fangxiaoyin on the expression of TLR7,IL-27,JAK2,STAT1 protein and mRNA in human bronchial epithelial cells stimulated by IL-4,and explore its possible internal mechanism for preventing and treating asthma in the remission period.Methods:Determination of the maximum non-toxic concentration of Gubenfangfangyin medicated serum on 16HBE cells by CCK8 method.A 16HBE cell model induced by IL-4 was established and randomly divided into a blank group,a model group,a montelukast group,a low-dose Gubenfangfangyin group,a medium-dose Gubenfangfangyin group,and a high-dose group.The expression level of IL-27 in the cell supernatant of each group was detected by ELISA,the expression level of IL-27,JAK2,STAT1 mRNA in each group of cells was detected by qPCR method,and the TLR7,JAK2,STAT1 protein expression levels in each group of cells were detected by Westernblot method.Results:Compared with the blank group,the expression level of TLR7 protein in the model group was down-regulated,and the expression level of JAK2 and STAT1 protein was up-regulated(P<0.05);the expression level of IL-27 in the cell supernatant was up-regulated(P<0.01);the expression levels of IL-27,JAK2,and STAT1 mRNA in the cells increased(P<0.05).Conclusion:Gubenfangfangyin can up-regulate TLR7 and inhibit the expression of IL-27 and JAK2/STAT1,which may be an internal mechanism for its prevention and treatment of asthma.
