Objective: Through animal experiments,this project improved the effect mechanism of BQTQF on pathological injury and inflammatory stress of nasal mucosa in rats.Through the addition of pathway blockers(H89 and PDTC),the mechanism of BQTQF affecting the expression of downstream mucins(MUC5A and MUC5B)by regulating the tandem communication between c AMP-PKA-CREB and NF-κB pathway was studied.According to the the molecule AR mechanism of the intervention of BQTQF on the expression of mucin in allergic rhinitis,the effect mechanism of BQTQF on the prevention and treatment of allergic rhinitis is supplemented,which provides a scientific basis for the promotion of BQTQF.METHODS: Using animal experiment method,combined with the use of pathway inhibitors(H89,PDTC),80 rats were divided into blank group,model group,Chinese medicine group,Biyankang group,H89 group,PDTC group,H89+ Chinese medicine group,PDTC+ Chinese medicine group.The pathological damage of nasal mucosa of rats was observed by HE staining.The expression of NF-κBp65 was observed by immunohistochemistry.The changes of Ig E in rat serum were detected by ELISA.The levels of NF-κBp65,MUC5 A and MUC5 B in nasal mucosa were detected by Western blot.RT-PCR was used to detect the levels of MUC5 A and MUC5 B factors and the expressions of pathpath-related proteins CREB,p65 and CBP in the nasal mucosa of rats to verify the experimental hypothesis.Results:(1)HE staining results of nasal mucosa of rats with allergic rhinitis: ciliary abscission and lodgings,hyperplasia and dilation of small blood vessels,infiltration of a large number of inflammatory cells,significantly increased number of submucosal glands,and severe abscission of nasal mucosa in model group.THE cilium and nasal mucosa in BQTQF group were basically intact,inflammatory cell infiltration and glandular hyperplasia were significantly reduced.(2)When NF-κB p65 was used as the primary antibody,the immunohistochemical mean optical density showed that the positive result of NF-κB p65 was almost not expressed in the blank group,but the content of NF-κB p65 in the model group was significantly increased,and the difference was statistically significant compared with t HE blank group(P<0.05).After subsequent drug treatment,the content of Biyankang group and Buqi Tongqiao prescription group decreased significantly,compared with model group,P<0.05,the difference was statistically significant.There was no significant difference between PDTC group and PDTC+ Chinese medicine group(P>0.05).THE positive optical density of NF-κB p65 in H89 group and H89+ Chinese medicine group was significantly decreased after the combination of Chinese medicine,and the difference was statistically significant(P<0.05).(3)ELISA results: THE Ig E content in the peripheral blood of the rats with allergic rhinitis was changed,and the Ig E content in themodel group was significantly higher than that in the blank group,the difference was statistically significant(P<0.05);Compared with the model group,the content of Biyankang group and BQTQF group decreased significantly(P<0.05),and the difference was statistically significant.Compared with PDTC+ Chinese medicine group,the content of PDTC combined with Chinese medicine decreased(P<0.05),and the difference was statistically significant.Compared with H89+ Chinese medicine group,the content of H89 decreased significantly(P<0.05),and the difference was statistically significant.(4)Western blot results: WB results showed that the contents of p65,MUC5 A and MUC5 B in the nasal mucosa of rats with allergic rhinitis were significantly higher in the model group than in the blank group(P<0.05),and the difference was statistically significant.Compared with the model group,Biyankang and BQTQF groups had lower content(P<0.05),the difference was statistically significant.Compared with H89+ Chinese medicine group,the content of H89 group was decreased after the combination of Chinese medicine,the difference was significant(P<0.05),the difference was statistically significant;Compared with PDTC+ Chinese medicine group,the content of combined Chinese medicine group decreased significantly(P<0.05),and the difference was statistically significant.THE WB results of CREB protein showed that the content of model group was decreased by P<0.05 compared with t the blank group,and the difference was statistically significant.Compared with model group,the contents of Biyankang and Buqi Tongqiao Ig E in intervention group were significantly increased(P<0.05),and the difference was statistically significant.However,compared with H89+ Chinese medicine group,the content of H89 was significantly increased(P<0.05),and the difference was statistically significant.Compared with PDTC+ Chinese medicine group,the P>0.05 was not statistically significant.(5)RT-PCR results: THE results showed that the contents of p65,MUC5 A and MUC5 B in the nasal mucosa of rats with allergic rhinitis were changed.Compared with the blank group,the contents of each protein in the model group were significantly increased,and the difference was statistically significant(P<0.05).THE results of p65 and MUC5 A showed that although there was a difference between PDTC group and PDTC+ Chinese medicine group,the P>0.05 was not statistically significant.THE results MUC5 b showed that although there was a difference between PDTC group and PDTC+ TCM group was significant(P<0.05),and the difference was statistically significant.Conclusion:1.BuQiTongQiaoFang can alleviate the pathological damage in the nasal mucosa of rats with allergic rhinitis,reduce the expression of NF-κBp65,reduce the infiltration of eosinophils,inhibit glandular hyperplasia,and repair the damaged nasal mucosa.2.BuQiTongQiaoFang can reduce the Ig E content in peripheral blood of rats with allergic rhinitis,and the effect mechanism of BQTQF on alleviating clinical symptoms may be related to the reduction of serum Ig E content.3.BuQiTongQiaoFang can regulate the tandem relationship between the disorderly c AMP-PKA-CREB pathway and NF-κB pathway in rats with allergic rhinitis,rebalance the balance of the disorderly two pathways,relieve the expression of mucin in rats with allergic rhinitis,and play the effect of prevention and treatment of allergic rhinitis.
