| Sleep is an extremely important physiological function to maintain human life activities,and good sleep quality is an essential condition to ensure human physical and mental health.With the increasing pressure of survival in modern society,insomnia caused by irregular work and rest is gradually threatening people’s physical and mental health.It is urgent to pay attention to the sleep health of all mankind.TCM has a wealth of experience in the treatment of insomnia;however,different doctors have different prescriptions,and it is difficult to form a unified theoretical system that can be directly inherited.Based on the data of 1142 clinical prescriptions in a tertiary hospital in Zhejiang Province from 2014 to 2019,this article uses data mining and network pharmacology analysis methods to explore the prescription and prescription rules of TCM for the treatment of insomnia,and the pharmacological mechanism of high-frequency Chinese medicine/drug group.The results can provide references for the development of Chinese medicine for the treatment of insomnia and the development of high-efficiency and low-toxic side western medicines,and provide a theoretical basis for the modernization of Chinese medicine.The following is the main content of this article:1.The application of data mining and network pharmacology analysis technology in the research of TCM prescriptions is introduced in detail.2.The data of 1142 prescriptions were analyzed.It is found that most of the prescription drugs for insomnia are mainly to soothe the nerves and tonic,supplemented by regulating qi,clearing heat,and relieving the surface;The medicinal properties are mainly Wenping,the medicinal taste is mainly sweet,the main meridian is the liver,and the spleen,heart,kidney,lung,and stomach meridians are second.These drugs improve the quality of sleep by regulating the balance of the five internal organs.3.Use Apriori algorithm model and systematic clustering to analyze commonly used prescription drugs.Obtained significant correlation of two groups,three groups,four groups of 11,10,4,and 5 clusters of new prescriptions.4.The pharmacological and pharmacodynamic mechanism of 8 high-frequency Chinese medicines and 4 groups of medicines were studied.The results show that 77 ingredients in 8Chinese medicines are effective for the treatment of insomnia.It mainly acts on 40 targets that are directly related to sleep rhythm regulation,such as SLC6A4,and 27 targets that have no direct effect,such as ADORA1.It mainly mediates 5-HT synapses,THs,Estrogens.,PI3K-Akt,DA synapses,cAMP,Tyr metabolism and other signaling pathways to regulate the synthesis and transport of related neurotransmitters,hormones,and regulate various neurotransmitters.Qualitative receptors,conduction axon activity,etc.So as to realize the regulation of human sleep rhythm,time to fall asleep,and wake cycle,and the mechanism of drug effect is clear. |
