| Purpose:(1)To evaluate the pharmacodynamic mechanism of Xiangefang on bone tissue.(2)By observing the effect of Xiangefang on TGF-β1/Smads signal pathway,we try to reveal its mechanism of intervention in PMOP.Methods:(1)In this experiment,ovariectomized rats were used as model group,and Xiangefang was used as experimental group.Rats undergoing sham operation were used as negative control.Alendronate sodium was given as positive control.(2)Bone mass of rats in each group was observed and analyzed with bone density as an index.Selecting blood Ca,p,ALP and StrACP as bone metabolism markers,measuring and analyzing serum data of rats in each group by using test box colorimetry,and observing bone metabolism of rats in each group.(3)Taking osteoblast proliferation as the study phenotype;The signal transduction of TGF-β1/Smads in bone tissue cells was taken as the research pathway.The mRNA and protein expressions of upstream factor TGF-β1 and downstream protein phosphorylated Smad2/3 in this signaling pathway were measured and analyzed by q-PCR and western blot,and the effects of xiangefang on signaling pathway were analyzed.(4)All data were collected and processed by SPSS statistical software version 26.0.Results:(1)The SD rats were modeled by ovariectomy.After 30 days of feeding,the bone mineral density,trabecular thickness and bone volume fraction were measured.The results showed that the negative control group was much higher than the other three groups,indicating the successful modeling.After 30 days of feeding with Xiange Decoction and alendronate sodium respectively,the bone mineral density,trabecular thickness and bone volume fraction of the administration group and the positive control group were higher than those of the model group and before administration.(2)The results of the administration group and the positive control group indicated that Xiange Decoction could promote the absorption of calcium and phosphate,and inhibit the activities of ALP and StrACP.(3)The mRNA and protein expressions of TGF-β1 and Smad2 in the administration group and positive control group were higher than those in the model group.Conclusion:(1)The effective mechanism of Xiange Decoction in treating PMOP may be that it regulates the expression of TGF-β1/Smads signaling pathway and promotes the proliferation and differentiation of osteoblasts,thereby maintaining the bone metabolism.(2)Xian Ge Fang can effectively relieve symptoms and improve general conditions of patients when used clinically.It can effectively improve the bone density of patients and promote the recovery of their trabecular structure and function.It can promote the increase of bone mineral content,enhance the anti-fracture ability,improve the state of bone tissue of patients,and regulate and stabilize the dynamic balance between bone absorption and bone formation. |
