Objective:Observe the influence of Suzidaotan Decoction on the airway remodeling of asthmatic mice,and investigate whether its mechanism is related to the wnt5 a signaling pathway.Methods:50 clean-grade female BALB/c mice were randomly divided into normal group,asthma mouse model group,Chaoyifang Suzidaotantang low-dose group,Chaoyifang Suzidaotantang high-dose group,and western medicine Dexamethasone positive control group.The other four groups except the normal group were sensitized on the first 1,7,14,and 15 days(to enhance the sensitization effect).After the sensitization,aerosol excitation was started for a total of 8 weeks,3 times a week.Gavage was performed at the same time as the aerosol stimulation.Except for the normal group and the model group,the normal saline was used instead of the drug.The other three treatment groups were treated with the corresponding dose and concentration of drugs: the Suzidaotan Decoction high-dose group was 200 μL/head,the concentration was 300 mg/kg;Suzidaotan Decoction low-dose group 200μL/mouse at a concentration of 150 mg/kg;western medicine dexamethasone group200 μL/mouse at a concentration of 0.5 mg/kg,once a day for 8 weeks.One hour after the last challenge,the mice were immediately sacrificed.Take the BALF,left lung tissue,and right lung tissue of the mouse,and store them in the appropriate place for storage for experimental use.The left lung tissues were sliced,and HE and Masson staining methods were used to study lung tissue pathological changes;acetylcholine was used to detect airway resistance in mice;ELISA method was used to detect asthma-related cytokines;IH and Western were used respectively The Blot method was used to detect the protein content of α-SMA and the phosphorylated protein expression of wnt5 a and NF-κB in the right lung tissue.Results:1.BALF results: Compared with the normal group,the BALF of the model group contains a large number of inflammatory cells;compared with the model group,it is found that the total number of BALF inflammatory cells and the classification count in the other three treatment groups have decreased,and each treatment group Compared with the model group,the improvement was obvious(P<0.05).2.ELISA test results: Compared with the normal group,the levels of cytokines(IL-4,IL-5,IL-13)in the BALF of the model group increased to varying degrees,and at the same time IFN-γ decreased(P<0.05);Compared with the model group,the contents of cytokines(IL-4,IL-5,IL-13)in the other three groups of BALF decreased compared with the model group,while the content of IFN-γ increased(P<0.05),the improvement was obvious.3.HE staining results: Compared with the normal group,the bronchial mucosa of the model group has a large area of ??inflammatory cell infiltration,and there is lung damage such as damaged alveoli(P<0.05);compared with the model group,it is found that the other three groups are treated The lung injury in the group improved to a corresponding degree,the improvement was obvious(P<0.05).4.Improved Masson’s trichrome staining results: Compared with the normal group,the model group had large areas of collagen fibers and airway smooth muscle thickening(P<0.05);compared with the model group,it was found that the other three treatment groups The area of ??blue collagen fibers and the thickening of the airway were reduced accordingly(P<0.05),the improvement was obvious.5.Measurement results of airway hyperresponsiveness: Compared with the normal group,the airway resistance of the model group was significantly increased under the action of different concentrations of acetylcholine(P<0.05);compared with the model group,it was found that the rest In the three treatment groups,airway resistance decreased to a certain extent under drug intervention,the improvement was obvious(P<0.05).6.IH results: Compared with the normal group,a large number of cells in the section of the model group were stained brown(P<0.05);compared with the model group,it was found that the yellow-brown cells in the other treatment groups were different in color(P<0.05).7.Western Blot results: Compared with the normal group,the protein content of α-SMA in the model group increased significantly(P<0.05);compared with the model group,it was found that the protein content of the other three treatment groups showed a decreasing trend(P<0.05).Compared with the normal group,the expression of wnt5 a and phosphorylated NF-κBp65 protein in the model group increased to a certain extent(P<0.05);compared with the model group,it was found that the protein expression levels of the other three treatment groups decreased,Showing a decreasing trend(P<0.05).Conclusions : The Chinese medicine prescription Suzidaotan Decoction can improve the effect of airway remodeling in asthmatic mice,and its mechanism of action is related to the wnt5 a signaling pathway.
