Objective: To replicate the animal model of TNBS ulcerative colitis,divide it into normal group,model group,Western Medicine Group and intestinal disease in different dosage treatment group,by observing the expression change of LPS-TLR4 axis,to explore the mechanism of immune regulation of intestinal diseases in ulcerative colitis,and to find a new target for the treatment of ulcerative colitis by TCM.Methods: 72 male SD rats with healthy cleaning grade,weight 145~150,randomly divided into 6 groups,each group of Animals 10: Normal group(group A),TNBS model group of Ulcerative colitis(group B),Positive Drug group(group C),Chinese Medicine bowel Disease Square Low Dose Group(group D),Chinese herbal bowel disease in the middle dose Group(group E),Chinese herbal bowel disease Square High Dose Group(group F).Feed normally for a week,give free food,water,and ensure feeding conditions.After a week to check the health status of SD rats,no special cases,in addition to the normal group continue to feed normally,do not mold,the remaining five groups are using TNBS induced colitis SD rat model.Continuous mold 3 days,after the end of 3 days of adaptive feeding,the 4th day began each group to feed the stomach 10 days: A,group B respectively to the drug of the same volume of saline,group C to 0.2g/kg the United States Mesalazine,the remaining three groups according to the "Lin academic experience set" to record the drug composition of the bowel disease side,Preparation of different concentrations of intestinal disease square extract(group D to the bowel disease 2.1G/kg,e group to the bowel disease 4.2G/kg,f group to the bowel disease side 8.4g/kg).After the end of the irrigation,the fasting 12 h,after anesthesia to collect specimens,observe the index.Results: 1.Compared with the normal group,the pathological score of colon tissue in the module increased significantly(p<0.05).Compared with the model group,the number of ulcers decreased,the infiltration of inflammatory cells decreased,the number of glands increased,and the pathological score decreased under the colon tissue microscope of TCM Group and control group.2.Compared with the control group,there was no significant change in the levels of LPS,IL-1 b,IL-6 and TNF-α in the model group.However,compared with the positive group of Western medicine,the expression of IL-22 in the model group increased(p<0.05)and the expression of IL-10decreased(p<0.05).3.Compared with the control group,the expression of IL-10 in the model group decreased,and a large number of IL-22 were expressed in various layers of the intestinal wall,and intestinal diseases inhibited the expression of IL-22 in the colon tissue of SD rats with ulcerative colitis.4.Compared with the control group,the expression of IL-10 in the model group decreased significantly(p<0.05),and the expression of IL-10 increased(p<0.05)after treatment with American salad.5.Compared with the control group,the expression of IL-22 in the model group increased significantly(p<0.05),and the expression of IL-22 decreased(p<0.05)after treatment with American salad.The expression of IL-22 in traditional Chinese medicine group decreased significantly(p<0.05),which was consistent with the results of immunohistochemical test.Conclusion: 1.Intestinal disease can treat ulcerative colitis induced by TNBS,and the optimum therapeutic drug concentration is medium and high dose concentration of traditional Chinese medicine.2.Intestinal disease can slightly reduce the LPS concentration of ulcerative colitis in rats,but it is not significant,and the expression of inflammatory factors such as IL-1β,IL-6 and TNF-α is not obvious.3.Intestinal diseases can significantly reduce the expression of IL-22,reducing the expression of TLR4 in diseased colon tissue.4.Intestinal diseases can increase the expression of protective cytokines IL-10.
