| Citri Reticulatae Pericarpium is the dried and mature peel of the rutaceae plant Citrus reticulata Blanco and its cultivated varieties.It is often used to treat abdominal distension,less food,vomiting and diarrhea,and more cough and sputum.The 2020 Chinese pharmacopoeia for tangerine peel has a single test index,and the content determination item only detects hesperidin,which is difficult to reflect the characteristics of tangerine peel polyflavonoids.Therefore,it is necessary to conduct a multi-index quality control study on Citri Reticulatae Pericarpium.Multi-index determination requires multiple reference substances,and reference substances are expensive and unstable in nature.This difficulty has not been solved.The method of one measurement and multiple evaluation has gradually attracted attention;however,in practice,conditions such as chromatographic columns need to be limited and important With low availability and poor accuracy,a new qualitative method needs to be established.In the first part of this thesis,the UPLC-Q-TOF/MS combined technology was used to systematically study the chemical composition of tangerine peel.The chemical composition of tangerine peel is very complicated.The development of a method that can quickly analyze and characterize the chemical components contained in tangerine peel is of great significance for a comprehensive understanding of the effective material basis of tangerine peel,the selection of quality control index components,and the identification of fingerprint chromatographic peaks.In the second part of this thesis,the quality control of 6 flavonoids in Citri Reticulatae Pericarpium is carried out by the quantitative analysis of multi-components by single-marker(QAMS).First,the chromatographic conditions suitable for the determination of Citri Reticulatae Pericarpium content were established,and the detection wavelength was investigated;secondly,the conditions such as the extraction method,extraction solvent,and extraction time of Citri Reticulatae Pericarpium were investigated to determine the optimal processing method for the test product of tangerine peel;Finally,the relative retention time value and relative correction factor of the remaining components to be tested were established by using hesperidin in tangerine peel as the internal reference.Qualitatively,the relative retention time value is relatively stable on the 5 chromatographic columns,and quantitatively,the relative retention time value is relatively stable.There is no significant difference between the calibration factor method and the external standard method.The last part of this thesis is the qualitative study of the eight flavonoids chromatographic peaks in Citri Reticulatae Pericarpium by the linear calibration using two reference substances(LCTRS)with the digital reference material software.First,according to the principle and operation method of DRS Origin software,a double-standard linear calibration method template for 8 kinds of flavonoids in tangerine peel was established,and 14 columns suitable for establishing the method were screened out from 25 chromatographic columns.Secondly,according to the results of the relative retention time method and LCTRS determined by the software,the two comparison methods are used to know that the double-standard linear correction method has ad vantages.Finally,the unknown chromatographic column was selected to verify the two qualitative methods,and the results showed that the secondary chromatographic column only met the method established by the LCTRS. |
