Objectives:To investigate the effects of artesunate and 5-FU on the proliferation,migration,apoptosis and cell cycle of human gallbladder cancer cells,and to compare the effect of the two drugs on cells,so as to explore the application prospect of artesunate in the treatment of gallbladder cancer.Methods:The OD values of artesunate and 5-FU were determined by CCK-8 assay and the IC50 values were calculated.The effects of artesunate and 5-FU on cell proliferation were observed by EdU-488 assay and clone formation assay.Cell scratches were used to detect the effects of artesunate and 5-FU alone or in combination on cell migration.Cells were labeled with Annexin V and PI double labeling,and apoptosis induced by artesunate and 5-FU alone or in combination was detected by flow cytometry.The cells were labeled with PI single label,and the effect of artesunate and 5-FU single and combined on the cell cycle was detected by flow cytometry.Results:CCK-8 method showed that the IC50 value of artesunate calculated from 48h OD data was 43.7μmol/L,and the IC50 value of 5-FU on cells after 48h treatment was 28.1 μmol/L.At 24h and 48h observation time points,compared with the control group,the 5-FU and artesunate groups significantly inhibited the proliferation of cells.EdU-488 method compared with the control group,the 5-FU group,artesunate group and the two-drug combination group significantly inhibited the proliferation of cells.The cell scratch results showed that compared with the control group,the 5-FU group,artesunate group and the two-drug combination group had significant inhibitory effect on cell migration.Annexin V and PI double label assay of apoptosis induced by 5-FU and artesunate groups and induced by the combination of two drugs showed that the 5-FU and artesunate groups and the combination of two drugs significantly promoted apoptosis compared with the control group.The effect of single PI on cell cycle in artesunate group,5-FUgroup and two-drug combination group showed that compared with the control group,5-FU group,artesunate group and two-drug combination group significantly inhibited the S phase and G2/M phase.Conclusion(s):1.Artesunate can inhibit the proliferation and apoptosis of GBC-SD gallbladder carcinoma cells in vitro;2.In vitro,artesunate combined with 5-FU had better anti-tumor effects than artesunate alone;3.In vitro experiments,artesunate combined with 5-FU had better anti-tumor effect than 5-FU alone;4.In vitro experiments,the antitumor effect of artesunate was weaker than that of 5-FU.
