Objective:To explore the clinical evidence that MDA-7/IL-24 has a tumor suppressor effect in papillary thyroid carcinoma,and provide a theoretical basis for the future diagnosis and treatment of MDA-7/IL-24 in papillary thyroid carcinoma.Methods:MDA-7/IL-24 SiRNA was used to transfect human papillary thyroid carcinoma cells to knock down the expression of MDA-7/IL-24,and the Thomsen-Friedenreich monoclonal antibody(A78-G/A7)was used to recover after intervention cell adhesion,migration and invasion tests are performed for verification and comparison.After the approval of the hospital ethics committee and the signing of the informed consent with the patients,serum samples from normal volunteers,benign thyroid nodules and patients with papillary thyroid carcinoma and tumor tissue specimens from patients with papillary thyroid carcinoma were collected.ELISA was used to detect the expression of MDA-7/IL-24 in the serum and analyze the differences in the expression of serum MDA-7/IL-24 in different groups of people;the collected papillary thyroid carcinoma tumor tissue was constructed on a tissue chip and passed immunohistochemistry Methods The expression of MDA-7/IL-24 in tumor tissues was detected;subsequently,the expression of MDA-7/IL-24 in the serum and tumor tissues of patients with papillary thyroid carcinoma was compared with some clinical prognostic indicators(including whether there is Lymph node metastasis,whether it is a multifocal tumor,thyroglobulin expression level,thyroid-stimulating hormone expression level,tumor pathological length,etc.)for further correlation analysis.Results:(1)In the cell adhesion experiment,compared with the control group(0.704±0.031),the cell adhesion index(0.264±0.012)of the mAb A78-G/A7 treatment group decreased significantly,while the knockdown MDA-7/IL-24 expression group(0.549 ± 0.067)significantly weakened the adhesion inhibition ability of mAb A78-G/A7,and the difference was statistically significant(P<0.05);in the invasion experiment,compared to the control group(585.700± 15.680),mAb The invasion ability of cells(146.700 ± 5.696)in the A78-G/A7 treatment group was significantly reduced,while the MDA-7/IL-24 SiRNA knockdown group(377.000±20.110)significantly reduced the ability of mAb A78-G/A7 to inhibit cell invasion,The difference was statistically significant(P<0.05);in the migration experiment,compared with the control group(704.000±23.440),the mAb A78-G/A7 treatment group(372.300 ±8.293)cell migration ability was significantly reduced,while The MDA-7/IL-24 SiRNA knockdown group(503.700±13.780)also weakened the ability of mAb A78-G/A7 to inhibit cell migration,and the difference was statistically significant(P<0.05).(2)Patients with benign thyroid nodules(372.800±51.350)and papillary cancer patients(stage I:375.300 ± 25.680;stage II/III:336.200 ±19.44)serum MDA-7/IL-24 concentrations are higher than those of healthy people(602.000)± 40.780)significantly decreased(P<0.01),but benign thyroid nodules were not significantly different from PTC patients,and there was no significant difference between the stages of papillary cancer patients;serum MDA-7/IL-in patients with papillary thyroid cancer There is no significant correlation between the concentration of 24 and whether there is lymph node metastasis,whether it is a multifocal tumor,the expression level of thyroglobulin,the expression level of thyroid-stimulating hormone,and the pathological length of the tumor;the results of tissue chip immunohistochemical staining indicate the presence of lymph node metastasis The expression level of MDA-7/IL-24 in the tumor tissue of patients with papillary thyroid carcinoma was significantly lower than that in patients without lymph node metastasis(P<0.05),but the expression level of MDA-7/IL-24 in tumor tissue was related to the multifocal and pathological tumor There was no significant correlation between the long diameter,the expression level of thyroglobulin and the expression level of thyroid-stimulating hormone.Conclusions:(1)Interference with MDA-7/IL-24 can attenuate the tumor suppressor effect of Thomsen-Friedenreich antibody in papillary thyroid carcinoma cells.Thomsen-Friedenreich antibody may exert its effect in papillary thyroid carcinoma cells by regulating MDA-7/IL-24;(2)The serum concentration of MDA-7/IL-24 shows low expression in papillary thyroid carcinoma,which is consistent with its low or no expression in other tumors,but MDA-7/IL-24 Serum concentration cannot judge the prognosis of papillary thyroid carcinoma;(3)Compared with the tumor tissue of papillary thyroid carcinoma with lymph node metastasis,the tumor tissue without metastasis shows high expression of MDA-7/IL-24,which is inferred to be papillary thyroid carcinoma.The expression of MDA-7/IL-24 in tumor tissues can be used as an indirect evidence reflecting the occurrence of lymph node metastasis.
