OBJECTIVE:Immunohistochemical method was used to detect the expression of SHP-1 and HMGB1 in breast cancer before and after neoadjuvant chemotherapy.The correlation between the expression of SHP-1 and HMGB1 before and after neoadjuvant chemotherapy and the clinicopathological characteristics was discussed.The relationship between the efficacy of neoadjuvant chemotherapy and the differential expression of SHP-1 and HMGB1 was explored.The correlation between the expression of SHP-1 and HMGB1 before and after neoadjuvant chemotherapy was analyzed.Methods:From November 2015 to November 2020,the clinical and pathological data of patients with breast invasive ductal carcinoma confirmed by pathology in the Second Affiliated Hospital of Nanhua University and the second people’s Hospital of Shenzhen City were collected.Among them,82 cases received neoadjuvant chemotherapy,82cases received coarse needle biopsy before neoadjuvant chemotherapy as group A,and 82 cases received postoperative biopsy as group B,The expressions of SHP-1 and HMGB1 were detected by immunohistochemistry;Objective to investigate the relationship between SHP-1,HMGB1 and clinicopathological features of breast cancer before and after neoadjuvant chemotherapy,analyze the expression differences of SHP-1,HMGB1 in breast cancer tissues before and after neoadjuvant chemotherapy,and evaluate the relationship between them and the efficacy of neoadjuvant chemotherapy.SPSS 26.0 software was used to analyze the data.Results:1.The positive expression rate of SHP-1 protein in group A was 45.7%(37/82),and the positive expression rate of SHP-1 protein in group B was 62.4%(51/82).The positive expression rate of SHP-1 protein in group B was higher than that before neoadjuvant chemotherapy,and the difference was statistically significant(χ~2=4.806,P=0.028<0.05).2.The positive expression rate of HMGB1 protein in group A was 76.5%(63/82),and the positive expression rate of HMGB1 protein in group B was 31.7%(26/82).The positive expression rate of HMGB1protein in group B was lower than that in group A,and the difference was significant(χ~2=33.635,P=0.000<0.05).3.The expression of SHP-1 protein in group A was related to the expression of ER(χ~2=9.288,P=0.002<0.05),PR(χ~2=37.841,P=0.000<0.05),and the expression of T stage before chemotherapy(χ~2=7.552,P=0.023<0.05),TNM staging(χ~2=7.552,P=0.023<0.05),HER-2 expression(χ~2=7.757,P=0.005<0.05),the difference was statistically significant.4.The expression of HMGB1 in group A was related to histological grade(χ~2=5.236,P=0.034<0.05),T stage before chemotherapy(χ~2=6.658,P=0.036<0.05),lymph node metastasis(χ~2=33.375,P=0.000<0.05)and TNM stage(χ~2=6.658,P=0.036<0.05),and the difference was statistically significant(P<0.05)5.The expression of SHP-1 and HMGB1 protein in the group before neoadjuvant chemotherapy was negatively correlated(r=-0.315,P<0.05),and the expression of SHP-1 and HMGB1 protein in the group after neoadjuvant chemotherapy was also negatively correlated(r=-0.334,P<0.05).6.Compared with the chemotherapy effect of SHP-1 negative patients in group B,the chemotherapy effect of SHP-1 positive patients was better(χ~2=8.271,P=0.004<0.05),the difference was statistically significant;Compared with the chemotherapy effect of HMGB1 positive patients in group B,the chemotherapy effect of HMGB1 negative patients was better(χ~2=7.934,P=0.005<0.05),the difference was statistically significant.Conclusions:The expressions of SHP-1 and HMGB1 proteins before and after neoadjuvant chemotherapy for breast cancer are different,showing a negative correlation,and are related to the efficacy of chemotherapy,suggesting that SHP-1 and HMGB1 proteins may have certain clinical value in predicting the efficacy of neoadjuvant chemotherapy for breast cancer.
