Objective:To explore and establish a method of comprehensive evaluation for the quality of Alpinia oxyphyllaFructs(AOF)associated with antioxidant activity.Method:(1)HPLC method was used to establish the fingerprint of AOF and UPLC-MS or reference material comparison method was used to identify part of the chromatographic peak structure.(2)Evaluating the antioxidant activity by taking advantage of methods of scavenging DPPH,OH· free radical and FRAP assays.(3)Active characteristic peak of scavenging DPPHfree radical by AOF was picked out by method of DPPH-HPLC.(4)Pearson correlation was used to analyze the relationship of "Spectrum-effect" of Antioxidant Activity of AOF and identify the characteristic peak of antioxidant activity.(5)The contents of 5-hydroxymethylfurfural,protocatechuic acid,protocatechualdehyde,chrysin and nootkatone extracted from AOF was determined by HPLC.(6)Combined with TOPSIS method the comprehensive evaluation results of the quality evaluation of AOF were obtained.Result:(1)HPLC fingerprints of AOFfrom 20 batches were developed and 23 common peaks were identified with the similarity between each sample and control fingerprint was above 0.963.The chemical structures of 11 chromatographic peaks in the fingerprint were identified,5 of which were common peaks.(2)The result of antioxidant experiment in vitro revealed that AOF have certain antioxidant activity by DPPH,OH· free radical scavenging and FRAP assays.The value of IC50 or ferric reduction equivalence is showed as follows:0.274~1.892 mg/mL、2.833~9.766 mg/mL 和 1.113~3.659 mmol/L.(3)Seven chromatographic peaks with DPPH radical scavenging activity were selected from the sample,which were protocatechuic acid,protocatechualdehyde,rutin,quercetin 3-galacturonide,kaempferol 7-glucuronide,4-Methoxybenzaldehyde and tectochrysin.(4)8 chromatographic peaks of antioxidant activity were screened by Pearson correlation analysis(P<0.05 or P<0.01),and the correlation coefficients were 0.468~0.752.(5)The content ranges of 5-hydroxymethylfurfural,protocatechuic acid,protocatechualdehyde,tectochrysin,and nootkatone detected by HPLC in AOF were as followed:0.00~103.92 μg/g,0.00~133.01μg/g,101.38~295.55 μg/g,80.98~157.79 μg/g,and 724.78~1675.76 μg/g.(6)TOPSIS method analysis results showed that when the antioxidant activity was used as the evaluation standard,European similarity ranking of 20 batches of AOF:S17>S12>S10>S2>S15>S19>S3>S11>S16>S7>S13>S20>S6>S9>S1>S8>S4>S18>S5>S14.Conclusion:It is the first time to use the comprehensive evaluation mode of spectrum effect relationship combined with TOPSIS method to obtain the results of quality evaluation of AOF,which can provide a scientific basis for more reasonable control and evaluation of the quality of AOF,and give some new ideas for exploring the quality control and evaluation methods that are more suitable for the characteristics of traditional Chinese medicine.
