| Objective:To investigate the expression and relationship of PD-1,PD-L1 and TP63 in plasma and cancer tissue of Esophageal Squamous Cell Carcinoma(ESCC),and study its correlation with pathological features and prognosis of patients.Methods:This study selected 72 cases with ESCC and 40 healthy controls from March 2019 to February 2020 in Nanchong Central Hospital.The preoperative plasma of 49 patients with ESCC and 40 healthy volunteers were collected,and collected 72 ESCC tissue specimens after surgery.The expression levels of PD-1,PD-L1,TP63 in peripheral blood and tissues were detected by enzyme linked immunosorb-ent assay(ELISA)and immunohistochemistry(IHC).Using SPSS 19.0 software processed the data,Kaplan-Meier analyzed the prognosis,to explore the expression and significance of PD-1,PD-L1 and TP63,P<0.05 was considered as statistical significance.Result:1.The plasma levels of sPD-1,sPD-L1 and sTP63 in ESCC were higher than those in the normal group,and the difference was statistically significant(P<0.05).In ESCC,the abnormal expression of sPD-1 was correlated to the vasculature invasion(P<0.05),and sPD-L1 and sTP63 have no correlation with clinicopathological factors.2.In patients’ tissues of ESCC,the positive expression rates of PD-1,PD-L1,TP63 were respectively 40.3%(29/72);45.8%(33/72);80.6%(58/72).The abnormal expression of PD-1 in tissues was correlated to lymph node metastasis and maximum diameter of the tumor of ESCC(P<0.05);Tissue PD-L1 positive expression was correlated to pathological stage,lymph node metastasis,and vascular invasion(P<0.05);Tissue TP63 positive expression was correlated with to lymph node metastasis,maximum diameter of the tumor,T stage,pathological stage,vascular invasion and nerve invasion of ESCC(P<0.05).3.In 49 cases of ESCC,TP63 and PD-1 in peripheral blood and tissues are correlated(P<0.05);sTP63 in peripheral blood is correlated with sPD-L1(P<0.05);sPD-1,sPD-L1 and sTP63 levels in peripheral blood are no correlated with PD-1,PD-L1 and TP63 in tissues(P>0.05).4.As of December 31,2020,the DFS and OS of patients with tissue PD-L1 positive expressions are shorter(P<0.05);and PD-1 positive expressions mean short OS,but not correlated to DFS;the positive expression of TP63 in tissues was not statistically significant with DFS and OS(P>0.05);sPD-1,sPD-L1 and sTP63 had no statistical significance between high expression and OS and DFS(P>0.05).Conclusions:1.The levels of sPD-1,sPD-L1 and sTP63 in the peripheral blood of esophageal squamous cell carcinoma are elevated;it is related to the occurrence and development of esophageal squamous cell carcinoma.2.There is overexpression of TP63 in esophageal squamous cell carcinoma tissue,it has certain value in determining the malignant degree of esophageal squamous cell carcinoma.The high expression of PD-1 and PD-L1 in esophageal squamous cell carcinoma tissue means a poor prognosis,which can be used as an indicator to determine the malignancy and prognosis of esophageal squamous cell carcinoma.3.In ESCC,TP63 has a certain relationship with PD-1.The combined detection of sTP63,sPD-1,and sPD-L1 in the peripheral blood of esophageal squamous cell carcinoma is helpful to evaluate the immune regulation of esophageal squamous cell carcinoma;it is also helpful to understand the malignancy and prognosis of the disease,which is a clinical diagnosis,treatment,and provide reference for prognosis judgment and formulation of comprehensive treatment plan. |
