Part 1:Effect of PEBP gel on embryo implantation number after endometrial injury in rats.Objective:Poly(ethyleneglycol)-b-poly(L-phenylalanine)gel(PEBP gel for short)was used in the model of intrauterine adhesion in SD female rats.The effect of PEBP gel on the embryo implantation number after endometrial injury in rats was observed by observing the number of embryo implantation in rats after endometrial injury.Methods:18 female SD rats were randomly divided into three groups:control group(group A),PEBP group(group B)and Gong’ankang group(group C).Control group:6 rats without modeling,other treatments were the same;PEBP group:6 rats were treated by curettage and infection double injury method to make rat model of intrauterine adhesion.After modeling,65mg/ml PEBP gel 0.3ml was randomly injected into one side of uterus as group B1,and normal saline 0.3ml was injected into the other side as group B2.Gong’ankang group:6 rats were made uterine adhesion model by curettage and infection double injury method.Later,0.3ml of Gong’ankang was injected randomly into one side of the uterus as group C1,and 0.3ml of normal saline was injected into the other side as group C2.After modeling,each mouse was reared in a single cage.All female SD rats were mated with male rats in 2:1 cages on the 8th day of experiment.The number of embryo implantation was observed on the 14 th day of pregnancy.Results:1.Control group:the number of embryo implantation in the left side(group A1)and right side(group A2)of SD rats was similar,and the difference was not statistically significant(P>0.05);2.PEBP group:the number of embryo implantation on PEBP gel side(group B1)was significantly higher than that in contralateral group(group B2),and the difference was statistically significant(P<0.05);3.Gong’ankang group:there was no significant difference in the number of embryo implantation between the side of Gong’ankang(group C1)and the opposite side(group C2)(P>0.05);4.The number of embryo implantation in group B1 was significantly higher than that in group C1,and the difference was statistically significant(P<0.05).Conclusion:Curettage plus infection double injury method seriously affects the number of embryo implantation in SD rats.PEBP gel can increase the number of embryo implantation after endometrial injury in rats.Part 2:Effect of PEBP gel on preventing adhesion after endometrial injury in rats.Objective:Different concentrations of PEBP gel in rat models of intrauterine adhesions,observe the degree of endometrial fibrosis and the expression level of TGF-β1,preliminary study on the mechanism of PEBP gel for preventing intrauterine adhesion in IUA model rats.Methods:36 female SD rats were randomly divided into 6 groups(group A,B,C,D,E,F).The rat model of uterine adhesion was established by uterine curettage and infection.In A,B,C,D and E groups,SD rats were established the model by randomly making one side of uterus and the uterus injected with0.3ml of PEBP concentration of Omg/ml,8.125mg/ml,16.25mg/ml,32.5mg/ml,65mg/ml respectively.In group F,SD rats were established the model by randomly making one side of uterus and the uterus injected with0.3ml normal saline as control group.Each rat was fed in a single cage after modeling,and the rats were killed 14 days after modeling.The middle uterine tissue of SD rats was fixed,embedded and sectioned.HE staining was used to observe the healing of endometrium,Masson staining was used to observe the degree of endometrial fibrosis,and the expression of transforming growth factor-β1(TGF-β1)in endometrial tissue was detected by immunohistochemistry.Results:1.After SD rats were made,PEBP gel injected into the uterine cavity could significantly inhibit the fibrosis of endometrium,and the degree of fibrosis decreased with the increase of PEBP gel concentration.2.PEBP gel could inhibit the expression of TGF-β1 in endometrial tissue,and the expression level of TGF-β1 decreased with the increase of PEBP gel concentration.The expression level of TGF-β1 in the E group with the highest PEBP concentration was the lowest.Conclusion:PEBP gel inhibited the fibrosis induced by endometrial injury in rats.The degree of fibrosis in endometrium decreased with the increase of PEBP concentration in the gel.The expression level of TGF-β1 is related to the PEBP gel concentration and decreases with the increase of PEBP gel concentration.It indicated that PEBP gel could inhibit the proliferation of fibroblasts and prevent adhesion.
