| Objective:To investigate the correlation between Gadolinium ethoxybenzyl diethylenetriamine pentaacetic acid(Gd-EOB-DTPA)enhanced magnetic resonance imaging(MRI)T1 mapping and Ki67 and micro vessel density(MVD)for VX2 liver cancer in rabbit,in addition,to explore the possibility of noninvasive evaluation of the expression of Ki67 and MVD in hepatocellular carcinoma(HCC).Methods:12 of rabbit VX2 liver cancer models were established by CT guided percutaneous seeding method.The enhanced MRI T1 mapping scan was performed by Gd-EOB-DTPA.50 of tumor regions of interest(ROI)were selected according to MRI images,and the experimental rabbits were sacrificed immediately.The liver was dissected and the expression of Ki67 and MVD in 50 ROI were detected.T1 relaxation time(Tlrt)of ROI was measured,including T1rt before enhancement(T1rt-pre),T1rt after enhancement(T1rt-post),Tlrt 20 minutes after enhancement(T1rt-20min),reduction rate of T1rt before and 20 minutes after enhancement(rrT1rt),rrT1rt=[(T1rt-pre minus T1rt-post)/T1rt-pre]×100%,the reduction rate of T1rt at 20 minutes after enhancement(rrTlrt-20min).Pearson correlation analysis of the correlation between Tlrt and expression of Ki67 and MVD.The T1rt between high and low expression groups of Ki67 and between high and low MVD groups was compared by independent sample t-test.Result:According to Pearson correlation analysis,Ki67 index was significantly positively correlated with Tlrt-pre and T1rt-20min;The correlation coefficients were 0.704 and 0.845 respectively(P<0.001).The Ki67 index was significantly negatively correlated with rrTlrt-20min,and the correlation coefficient was-0.779(P<0.001).MVD has a low degree of negative correlation with Tlrt-pre,with a correlation coefficient of-0.368(P<0.001);MVD has a moderate negative correlation with Tlrt-20min,with a correlation coefficient of-0.443(P<0.001);MVD has a low degree positive correlation with rrTlrt-20min,the correlation coefficient was 0.396(P<0.001).In the comparison of the Ki67 high expression group and the low expression group,Tlrt-pre and T1rt-20min were 1866.88±305.92(ms)vs.1530.23±251.38(ms)(P<0.001),and 1725.47±338.08(ms)vs.1229.80±278.24(ms)(P<0.001),respectively,all of them have significant difference(P<0.05).In the comparison of high and low MVD groups,T1rt-pre and T1rt-20min were 1163.70±266.89(ms)vs.1615.84±242.09(ms)(P<0.001),818.40±173.43(ms)vs.1341.60±295.63(ms),respectively,(P<0.001),all of them have significant difference(P<0.05).Conclusion:The T1rt of the enhanced MRI with Gd-EOB-DTPA is related to the expression of Ki67 and MVD.T1rt-20min has high diagnostic value for the high expression of Ki67 and high MVD in rabbit VX2 liver cancer.Objective:To evaluate the early effect of cryoablation of VX2 liver cancer in rabbits by Gd-EOB-DTPA enhanced MRI T1 mapping,and to provide the theoretical basis for follow-up of Gd-EOB-DTPA enhanced MRI after cryoablation of HCC.Methods:20 residual tumors of rabbit VX2 liver cancer models were prepared by cryoablation.The Gd-EOB-DTPA enhanced MRI T1 mapping was performed on 7 days after tumor cryoablation.The values of Tlrt-pre,T1rt-20min and apparent diffusion coefficient(ADC)of residual lesions and necrotic lesions were measured.Independent sample t-test was used to compare Tlrt-pre,T1rt-20min and ADC between residual lesions and necrotic lesions.Receiver operating characteristic(ROC)curve was used to analyze the diagnostic value of Tlrt-pre,T1rt-20min and ADC for residual tumor.Result:The T1rt-pre,T1rt-20min and ADC in the rabbit residual tumor lesion and necrotic lesion after cryoablation were 1644.77±455.96 msvs.1041.69±318.26 ms(P<0.001),1327.96±425.59 ms vs.574.00±159.83 ms(P<0.001),1.19×10-3±0.21×10-3 mm2/s vs.1.50×10-3±0.25×10-3 mm2/s(P<0.001),respectively,all of them have significant difference(P<0.05).The sensitivity and specificity of Tlrt-pre,T1rt-20min and ADC in identifying residual tumor after cryoablation in tumor-bearing rabbits were 77.5%,85.0%(cut-off value 1347.8ms),87.5%,92.5%(cut-off value 814.7ms),77.5%,77.5%(cut-off value 3.35×10-3mm2/s),respectively.The AUC of T1rt-pre,T1rt-20min,and ADC were 0.854(P<0.001,95%confidence interval(CI):0.757,0.923),0.960(P<0.001,95%CI:0.891,0.991),0.842(P<0.001,95%CI:0.744,0.914),respectively.Conclusion:The Tlrt value of Gd-EOB-DTPA enhanced MRI is a valuable index for clinical application to evaluate the early effect of cryoablation of liver cancer.The sensitivity and specificity of T1rt-20min in the diagnosis of residual tumor after cryoablation were higher than those of Tlrt-pre and ADC. |
