| Objective:The paper is to detect the expression level of IL-8 in the issues of patients with triple negative breast cancer by immunohistochemical staining,and to analyze the relationship between IL-8 expression level and the clinicopathological characteristics,survival and prognosis of patients.Methods:The study collected the clinical data of 93 patients with triple negative breast cancer(TNBC)treated in the affiliated Hospital of North Sichuan Medical College from January 2010 to December 2018.According to the inclusion and exclusion criteria,a total of 80 patients were included in the experiment.At the same time,Paraffin specimens of these patients were collected to make tissue sections,and at the same time,the expression level of IL-8 in 80 TNBC patients was detected by immunohistochemical staining.The average positive area rate of IL-8 was calculated by Image J software.According to X-tile,the best cut-off value was obtained.And the patients were divided into IL-8 high expression group and low IL-8 expression group.Chi-square test was Firstly used to analyze the relationship between the expression level of IL-8 and the clinicopathological features of TNBC patients.Then the survival and prognosis of patients with high and low expression of IL-8 were analyzed by Kaplan-Meier method.Next the paper analyzed the difference of survival prognosis between patients with high expression of IL-8 and those with low expression of IL-8 by Log-rank test.Finally,the effect of many factors on disease free survival(DFS,disease free survival)was analyzed by Cox proportional hazard regression model.Results:Among the 80 TNBC patients,18 cases had high expression of IL-8,accounting for 22.5%of the total number of patients,and 60 cases of low expression accounted for 75%of the total number.Univariate analysis showed that the expression intensity of IL-8 in triple negative breast cancer was significantly different from clinical stage,Ki-67 expression,WHO grade,number of lymph node metastasis and tumor size(P<0.05),but not from age,pathological type and menopausal status(P>0.05).Secondly there was no significant difference between the cancer tissues of TNBC patients who had received neoadjuvant chemotherapy and those who had not received neoadjuvant chemotherapy(P>0.05).The results of Kaplan-Meier survival analysis showed that the median survival time of DFS in patients with high expression of IL-8 was lower than that in patients with low expression of IL-8(28 months VS 36 months).Log-rank test showed that there was a significant difference in DFS between the two groups.Through Cox proportional hazard regression model analysis,the results showed that the expression level of IL-8 was an independent factor affecting the survival and prognosis of TNBC patients(HR:1.15 5,95%CI:1.049-1.272,P=0.003).The prognosis of TNBC patients with high expression of IL-8 was poor.Conclusion:There is a difference in the expression of IL-8 in patients with triple negative breast cancer,and the expression intensity of IL-8 is an independent risk factor affecting the survival and prognosis of patients with TNBC. |
