Purpose:We designed to investigate the significance of Nucleophosmin(NPM)and proliferating cell nuclear antigen(PCNA)expression in non-small cell lung cancer tissues.We also evaluated the correlation between NPM and PCNA proteins expression and clinicopathological features of NSCLC patients,with an aim to determine their clinical significance in this malignant tumor.Materials and methods:Collected 42 cases of paraffin-embedded tissues and 36 cases of corresponding adjacent lung tissues from patients with NSCLC who underwent radical pneumonectomy from January 2018 to June 2019 in the Fourth Affiliated Hospital of XX University as the research objects.NPM and PCNA proteins expression in NSCLC and adjacent lung tissues were detected by immunohistochemical SP staining method.Single factor analysis was used to explore the correlation between NPM and PCNA proteins expression and clinicopathological features of NSCLC patients.Spearman rank correlation analysis evaluated the correlation between NPM and PCNA proteins expression.SPSS 25.0 statistical software package was used for statistical analysis,there was statistically significance in differences(p﹤0.05).Results:1 NPM and PCNA proteins expression in NSCLC tissues1.1 NPM protein expressionThe positive expression rate of NPM protein was 71.4%(30/42)in NSCLC tissues and 26.2%(11/42)in adjacent lung tissues.The positive expression rate of NPM protein in NSCLC tissues was significantly higher than that of adjacent lung tissues was statistically significant difference(p﹤0.05).1.2 PCNA protein expressionThe positive expression rate of PCNA protein was 88.1%(37/42)in NSCLC tissues and 19.0%(8/42)in adjacent lung tissues.The positive expression rate of PCNA protein in NSCLC tissues was significantly higher than that of adjacent lung tissues was statistically significant difference(p﹤0.05).2 The correlation between NPM,PCNA proteins expression and clinicopathological features in NSCLC2.1 The correlation between NPM protein expression and clinicopathological features in NSCLCAccording to the result of immunohistochemistry semi-quantitative integration method showed that the high and low expression of NPM protein are respectively 27(++,+++)and 15(﹣,+)in NSCLC tissues.The Mann-Whitney U test analysis showed that the level of NPM protein expression was negatively correlated with the differentiation of NSCLC was statistically significant difference(p﹤0.05).While the level of NPM protein expression was uncorrelated with age,gender,clinical stages,lymph gland metastasis,tumor size and pathological type of NSCLC patients was no statistically significant difference(p﹥0.05).2.2 The correlation between PCNA protein expression and clinicopathological features in NSCLCAccording to the result of immunohistochemistry semi-quantitative integration method showed the high and low expression of PCNA protein are respectively 32(++,+++)and 10(﹣,+)in NSCLC tissues.The Mann-Whitney U test analysis showed that the level of NPM protein expression was uncorrelation between degree of differentiation,age,gender,clinical stages,lymph gland metastasis,tumor size and pathological type in NSCLC(p﹥0.05).3 The correlation between NPM and PCNA protein expressionIn the identical NSCLC tissues,Among 32 cases of high expression of PCNA protein 26 cases of high expression of NPM protein,among 10cases of low expression of PCNA protein only 1 cases of high expression of NPM protein.Spearman rank correlation analysis showed that PCNA and NPM protein expression are positively correlated(r_s=0.633,p﹤0.05)。Conclusion:1 Both NPM and PCNA protein are highly expressed in NSCLC tissues indicating that both have a malignant progression effect in the occurrence of this tumor.2 The level of NPM protein expression was negatively correlated with the degree of differentiation,which is of great value to evaluate the malignant degree of NSCLC in clinical pathology.3 NPM and PCNA protein are positively correlated in the expression of NSCLC tissues indicating that NPM protein has a postive role in the proliferation of NSCLC.
