| Objective:To evaluate the simple effects and synergistic interactions of components of Weichangshu Tablets on gastric emptying and intestinal propulsion by factorial design method;to analyze the pharmacokinetics and tissue distribution of active ingredients of Weichangshu Tablets in rats and explore its absorption and distribution in the body,which laid a material foundation for the secondary development of Weichangshu tablets.Methods:1.SD rats were randomly divided into 8 groups,namely Weichangshu Tablet group(Radix paeoniae Alba+Rhubarb+Glycyrrhiza uralensis),Rhubarb and Radix paeoniae Alba group,Rhubarb and Glycyrrhiza group,Radix paeoniae Alba and Glycyrrhiza group,Radix paeoniae Alba group,Rhubarb group,Glycyrrhiza group,blank group.Each group was given Weichangshu tablets and prescription medicines for 7 days.With dextran blue(BD)2000 as the marker,the absorbance density value was measured at 620nm by an ultraviolet-visible spectrophotometer to obtain the gastric pigment emptying rate of each rat.The total intestinal distance and the intestinal propulsion distance of the BD2000 solution were measured to calculate the intestinal propulsion rate data.SPSS 20.0 statistical software was used for factorial design analysis of variance.2.SD rats were randomly divided into 9 groups,1 group was the blank group,and the rats in the blank group were given sterile saline.The other 8 groups were given Weichangshu solution,which was administered by 20 ml/kg/day,once a day,for 7 days.After the last administration,rats in each group were killed at 0.5、1、2、4、6、8、10、12 hours respectively.Thymus,spleen,heart,liver,kidney,stomach,intestine and brain tissues were dissected and collected immediately.HPLC-MS/MS was used to detect the active ingredients in different groups of rats and their concentrations in different time periods,and DAS 2.0 software was used to calculate the tissue distribution dynamic parameters.Results:1.Glycyrrhiza uralensis had the simple effect of promoting gastric emptying(p<0.05),and the compatibility of radix paeoniae alba,Rhubarb and Glycyrrhiza in Weichangshu Tablets had an interaction effect on promoting gastric emptying(p<0.05).Single-flavored radix paeoniae alba,rhubarb,and Glycyrrhiza had separate effects to promote intestinal propulsion(p<0.05),and the combination of rhubarb and radix paeoniae alba had the effect of promoting intestinal propulsion(p<0.05).2.After intragastric administration of Weichangshu tablets in rats,the distribution characteristics of each component are as follows:formononetin was highest in heart tissue samples at 2h and gradually decreased after 2h;emodin was highest in liver tissues at 8h;the concentration of rhein was highest in kidney tissue samples at 6h;the concentration of aloe-emodin in spleen tissue samples was highest at 1h;the concentration of rhein in gastric samples was highest at 1h;the concentration of glycyrrhizin in intestinal tissue products was highest at 0.5h;The concentrations of 6 compounds in thymus tissue samples were all low;the aloe-emodin concentration was the highest in brain tissue samples at 0.5 h.After 8 hours of administration,the tissue concentrations of the six components in each viscera decreased.Six compounds were detected between 0.5-2 h,indicating that the compounds could be rapidly distributed in the tissues after oral administration..Glycyrrhizin is absorbed more in the intestine,isoliquiritigenin,aloe-emodin and rhein are absorbed more in the stomach,which is consistent with the gastrointestinal motility promoting effect of Weichangshu Tablets.Conclusion:Weichangshu Tablets is scientific and its active ingredients have a wide distribution and absorption in various tissues.The tissue distribution results further confirm that Weichangshu Tablets is a typical Chinese medicine prescription for treating gastrointestinal insufficiency. |
