Objective:To observe the effect of Qingxiao method on the auricle inflammatory response in acne rats and to explore the mechanism of treatment of acne.Methods: Forty rats were randomly divided into blank group,model group,western medicine group and traditional Chinese medicine group,each group of 10,using the method of auricle injection of Propionibacterium acnes to copy the acne model.The traditional Chinese medicine group was given iso-vitaminic acid capsule,the other two groups were given the same dose of normal saline,gavage,once a day,after 21 days,observe the ear appearance and pathological changes of rats,measure the thickness of rat auricle,calculate the swelling rate of rat auricle tissue,enzyme-linked immunosorbent assay(ELISA)to detect TNF-α,IFN-γ content,immunohistochemical(IHC)and Western blot in rat serum ternblot detection of TLR-2,My D88,NF-κB,IL-1βprotein expression in rat focus tissues.Results:1.Changes of auricle appearance in ratsThe auricle of the rats in the blank group was soft,the color was pale pink,the capillaries were clearly visible,there were no papules pustules,pimples,horn bolts and other skin lesions;the ears of the rats in the model group were dark red,the hair follicle orifice was thickened,rough and dry,there were both papules and pustules,and the capillaries of the dermis were dilated obviously.2.Changes of auricle thickness and swelling rate in ratsBefore modeling,there was no significant difference in auricle thickness in each group(P>0.05).After modeling,the ear thickness and swelling rate of rats in the model group were significantly increased(P<0.05).Compared with the model group,the ear thickness and swelling rate of rats in the western medicine group and the traditional Chinese medicine group were improved(P<0.05).Compared with western medicine group,there was no significant difference in auricle thickness and swelling rate of rats in Chinese medicine group(P>0.05)3.Pathological changes of rat auricleThere was no obvious abnormality in auricle tissue,no inflammatory cells,and the boundary of each layer of epidermis was clear and complete.Filling and plugging extended to sebaceous glands,the funnel was enlarged in the form of kettle,and inflammatory cells were infiltrated around the hair follicles;the epidermis of western medicine group was thickened and relieved,the hair follicle keratosis was alleviated,only a small amount of inflammatory cells were infiltrated;the hair follicle dilatation degree of traditional Chinese medicine group was obviously alleviated,inflammatory cell infiltration was reduced,and the keratinization layer gradually returned to integrity.4.Serum TNF-α and IFN-γ in ratcompared with the blank group,the serum TNF-α content of rats in the model group increased and the content of IFN-γ decreased,the difference was statistically significant(P<0.05).compared with the model group,the serum TNF-α and IFN-γ content of rats was no significant difference in the traditional medicine group(P>0.05).5.The expression of TLR-2,My D88,NF-κB and IL-1β in rat auricle was detected by immunohistochemical methodCompared with the blank group,the expression of TLR-2,My D88,NF-κB,IL-1β in the auricle of the model group was up-regulated,the difference was statistically significant(P<0.05);compared with the model group,the expression of TLR-2,My D88,NF-κB,IL-1β in the auricle of the traditional Chinese medicine group and the western medicine group were significantly down regulated(P<0.05);but the difference between the traditional Chinese medicine group and the western medicine group was not statistically significant(P>0.05).6.The expression of TLR-2,My D88,NF-κB and IL-1β in rat auricle was detected by WesternblotCompared with the blank group,the expression of TLR-2,My D88,NF-κB,IL-1β in the auricle of the model group was up-regulated,the difference was statistically significant(P<0.05);compared with the model group,the expression of TLR-2,My D88,NF-κB,IL-1β in the auricle of the traditional Chinese medicine group and the western medicine group was down regulated,the difference was statistically significant(P<0.05);compared with the western medicine group,the expression of TLR-2 protein in the auricle of the traditional Chinese medicine group was down regulated,the difference was statistically significant(P<0.05)The expression of My D88,NF-κB,IL-1βprotein was not significant(P>0.05).Conclusions:1.Qingxiao method can improve the local inflammatory response of auricle in acne model rats.2.by modulating the TLR-2/My D88/NF-κB signaling pathway,qingxiao reduced the expression of TLR-2,My D88,NF-κB,IL-1β protein,reduced the serum TNF-α content,increased the content of IFN-γ and inhibited the local inflammatory response in the auricle of the acne model rats to achieve the effective treatment of acne.
