Study On The Quality Standard And Quality Evaluation Of Plantaginis Semen And Its Decoction Pieces

Plantaginis Semen is a common Chinese medicine commonly used in China.It is the dried mature seeds of Plantaginis Semen plant Plantago asiatica L.or Plantago depressa Willd.,mainly produced in Jiangxi,Heilongjiang,Liaoning,Hebei and other places.Plantaginis Semen is first recorded in "Shen Nong’s Herbal Classic".It contains many types of chemical components such as phenylethanol glycosides,cyclic olefin ether terpenes,flavones,sterols,and alkaloids,can treat chronic tracheitis,constipation,hypertension,gout and other diseases,direct use or broaching with salt water.At present,there are dozens of Chinese medicine preparations using Plantaginis Semen as the main raw material,so the market demand for Plantaginis Semen is very large.However,Plantaginis Semen is mature in stages and has a long harvest period,so it is often filled with fakes,mixed with fakes,and has a small particle size,deep color,and irregular shape,so it is difficult to distinguish the authenticity with the naked eye.The rough processing results in some stones and sediment mixed into the Plantaginis Semen,and the storage conditions are not up to standard,which leads to excessive water content,which seriously affects the efficacy and safety of Plantaginis Semen.The current quality standards of Plantaginis Semen are not very good.To control its quality and safety,so this project uses the Chinese Medicine Standardization Project of the State Administration of Traditional Chinese Medicine(ZYBZH-Y-JX-28)as the source of the project to carry out quality standards and quality evaluations of Plantaginis Semen officinalis and salt Plantaginis Semen pieces.The research aims to provide some scientific basis and method reference for the safety and quality control of Plantaginis Semen.This article takes Plantaginis Semen and its commonly used processed products,saltPlantaginis Semen,as the research object.First,the quality standards of Plantaginis Semen and medicinal herbs and salt Plantaginis Semen pieces are improved.Secondly,the process of processing salt Plantaginis Semen pieces is carried out.The classification and quality evaluation of the correlation between internal quality and appearance shape were studied by combining with multivariate statistical analysis,network pharmacology for screening of major pharmacological active components of Plantaginis Semen and analysis of pharmacodynamic mechanisms,and finally the quality markers of Plantaginis Semen are predicted to build a quality control and quality evaluation system with Plantaginis Semen as the core.The research content is as follows:(1)Study on the quality standards of Plantaginis Semen officinalis and salt Plantaginis Semen pieces: Based on the quality standards of Plantaginis Semen Plantaginis Semen in the 2015 edition of the Chinese Pharmacopoeia,48 batches of Plantaginis Semen seeds and 55 batches of salt Plantaginis Semen pieces were used as The research object carried out comprehensive quality standard research from thin-layer identification,microscopic identification,inspection items,content determination,heavy metal detection,aflatoxin detection,pesticide residues,etc.,and optimized the method of thin layer chromatography,added Isoacteoside index components under the content determination item,at the same time the fingerprint of Plantaginis Semen was established,and the components of the chromatographic peaks of the fingerprint chromatogram were identified by the combination of liquid phase and mass spectrometry.Meanwhile,the fingerprints were used to identify five mixed fakes such as Lepidii Semen and Celosiae Semen.(2)Study on processing technology of salt Plantaginis Semen: track the whole process of processing of Plantaginis Semen to salt Plantaginis Semen,formulate the production process parameters of salt Plantaginis Semen production,compare the composition changes during processing,and find that a part of the Acteoside components will be converted Isoacteoside under heating.(3)Quality classification of Plantaginis Semen and Salt Plantaginis Semen: use theprevious data samples,use a variety of statistical analysis methods,select indicators that can measure the quality characteristics of Plantaginis Semen,and analyze the correlation between external traits and internal quality.At the same time,combining the feasibility of market application,Plantaginis Semen medicinal materials and decoction pieces are divided into three grades: unified goods,first-class and second-class,in order to achieve efficient,accurate,and operable grade evaluation in the market circulation process.(4)Plantaginis Semen network pharmacology research and quality marker prediction: by searching the database and consulting the literature,screening Plantaginis Semen active ingredients,performing target prediction and drug-target network analysis,predicting and discussing from the molecular level the multi-component,multi-target overall regulation function of Plantaginis Semen provides reference for in-depth basic experimental research and clinical rational application of Plantaginis Semen.Finally,considering the affinity,effectiveness,and measurability of the chemical composition,the quality markers of Plantaginis Semen are predicted to Geniposidic acid,Acteoside,and Isoacteoside,which provide theoretical support for the quality standard and grade evaluation research in the early stage.
