| Asthma is one of the stubborn disease that jeopardizes the health of peoples around the world.It is a combination of multiple cells,inflammatory mediators and multiple targets.Repeated cough,expire,difficulty breathing,chest tightness and other clinical manifestations。Currently,There are more than 300 million people with asthma worldwide and asthma brings great burden to people’s life.Traditional Chinese medicine has a long history,rich theoretical foundation,and has clinical experience in the formulation of medicinal materials and its effectiveness in treating diseases and preventing diseases.However,there are many components,the diversity of therapeutic targets and The main chemical components of the traditional Chinese medicine compound have not been studied yet.Luoouke zupa is a folk compound widely used in xinjiang for the treatment of stubborn phlegm,asthma and other respiratory diseases,made from Hyssopus cuspidatus Boriss.And Iris halophila pall.This research group studied the anti-asthmatic effect of this compound and the chemical composition of the main medicinal herb Hyssopus cuspidatus Boriss.,clarified the anti-asthmatic effect,and had a preliminary understanding of the chemical composition of the main medicinal material.this study is based on our previous work,we further separated different eluting parts of the Luoouke zupa compound extract,screened out the best active parts,and analyzed the components of the root of the compound constituent Iris halophila pall.Separation of water,30%,50%,70%,90% ethanol elution site of Luoouke zupa extract by macroporous adsorption resin.The anti-asthmatic effect of the eluted parts of the extract of Luoouke zupa extract was studied in vivo using the rat model of asthma established by ovalbumin sensitization.After 15 days of intragastric administration,the lung tissue of rats was taken for HE staining to detect cytokines in serum and bronchoalveolar lavage fluid.The results of HE staining showed that the pathological changes of bronchial infiltration,alveolar wall thickening and hyperemia in the lung tissue of each treatment group were significantly reduced,and the secretion was significantly reduced.The results of serum cytokine assay showed that :The concentration of IL-4 in the water group was significantly decreased(P < 0.001),the concentration of IL-4 in the30%,90% ethanol group was significantly decreased(P < 0.05,P < 0.01),the concentration of IL-6 in the 50% ethanol group was significantly decreased(P < 0.01).The results of bronchoalveolar lavage fluid cytokine test showed that The level of IL-17 in bronchoalveolar lavage fluid of rats in the water site group was significantly decreased(P < 0.05).According to the symptoms of the rats in the model group and the test instructions,the model of bronchial asthma in rats was successfully established,and the site of Luoouke zupa in each group had certain therapeutic effect on asthma,among which the water site of Luoouke zupa had the best therapeutic effect.LC-ESI-Q-TOF-MS/MS method was used to analyze the chemical constituents of the medicinal materials Iris halophila pall.of the medicinal materials Luoouke zupa and18 chemical components were detected.it is:Tectorigenin A、Iris tectorigenin B1 或 Iris tectorigenin A1 、 7-O-methyl mangiferin 、 Irilin Da 、 Luteolin-6-C-β-D-glucoside 、Tectorigenin-7-O-glucosyl-4-O-glucoside 、 Iris tectorigenin B-7-O-β-glucosyl(1→6)glucoside 或 Iris tectorigenin A-7-O-β-glucosyl(1→6)-glucoside、3、-Hydorxytectoridin,Tectorigenin-4、-O-β-D-glucoside、Iristectorin A 或 Iristectorin B、Dihydrokaempferol-7-O-glucoside、Dalspinosin、Nigrigin、Irilin D、Iristectorigenin B、Isotectorigenin、Irilone a 和 6,4、-dimethoxy-5,7-dihydroxy-isoflavone。This result lays a foundation for further research on the basic research of pharmacodynamic components and the in-depth study of the sinusoidal appendix in the future.In summary,we have basically clarified the anti-asthmatic effect of the various elution sites of Luoouke zupa and have a certain understanding of the chemical composition of Iris halophila pall.,which is for future research.Learn from the meaning. |
