BackgroundPerchlorate(Cl O4-)is a water-soluble compound widely distributed in ecosystems[1].Perchlorate in the environment is extremely stable as an inorganic salt,and it persists in soil and groundwater for a long time,causing considerable controversy on human health.In recent decades,most of the research on perchlorate has been to explore the toxic effects of perchlorate on the thyroid domestically and overseas,and to study the potential correlation between perchlorate and thyroid function.And metabolism play a very important role[2-4].This article studies the effect of perchlorate on liver lipid metabolism in mice,which has certain significance for human health.MethodIn this study,48 male C57BL/6J mice were separate into four groups(n=12),and three groups(LG,MG,HG)of different doses of perchlorate aqueous solution(0.1mg/kg bw/d,1mg/kg bw/d,10mg/kg bw/d)fed mice,a group of blank control(CG)group was fed with distilled water to explore the liver lipid of C57BL/6J mice with different doses of perchlorate Metabolic effects.After male C57BL/6J mice were fed with conventional feed(14.25%protein,26.37%starch,1.11%fat)for 12 weeks,the mice were euthanized,the liver of the mice was quickly removed,weighed and the liver organ index At the same time,tissues of the same part of the liver were taken for oil red O staining section to observe the distribution of lipid droplets in the liver of mice.Determination of serum lipid metabolism indexes TG,TC,HDL-C and LDL-C and oxidative damage indexes MDA and GSH,as well as liver function damage indexes AST and ALT,liver tissue TG and TC content And the mice were tested for glucose tolerance and insulin tolerance.The content of fatty acid synthesis key rate-limiting enzymes ACC,FAS and fatty acid decomposition key rate-limiting enzyme CPT-1 in liver tissue was determined,and fatty acid composition and content of liver and adipose tissue were measured.Result1.After 12 weeks of exposure to perchlorate in mice,compared with the blank group(CG),the body weight of the HG(10mg/kg bw/d perchlorate intake)group had a significant difference(P<0.05).The results showed that a perchlorate intake of 10 mg/kg bw/d caused weight gain in mice.By comparing the organ index of the blank control group,it can be found that 10mg/kg bw/d perchlorate intake will increase the liver organ index of mice and is significant(p<0.05).The results showed that a perchlorate intake of 10 mg/kg bw/d caused lipid accumulation in the body and increased liver organ index in mice.Observed by oil red O staining of mouse liver tissue,after 12 weeks of ingestion of 10 mg/kg bw/d perchlorate in mice,the oil red O staining solution was stained with red lipid droplets surrounded by cell membranes.More lipid accumulation.The results showed that a perchlorate intake of 10 mg/kg bw/d caused lipid accumulation in the liver of mice.2.After 12 weeks of exposure to perchlorate,the mice were found by studying blood lipid levels,using the blank control group as a reference,by measuring serum TC,TG,LDL-C,HDL-C content,liver function indexes AST and ALT content,The content of oxidative damage indicators GSH and MDA,the content of TG and TC in mouse liver tissue,glucose tolerance and insulin tolerance,the results show that the perchlorate high dose group(intake of 10mg/kg bw/d)will affect The fatty acid metabolism in the liver of mice is disturbed,and the effect of perchlorate on fatty acid metabolism in the liver of mice is related to its dose.Enzymes and genes related to fatty acid metabolism in mouse liver are disordered,which affects the synthesis and decomposition of fatty acids.3.By measuring the content and composition of fatty acids in the peri-renal fat of mice,the results showed that the content of saturated fatty acids in the peri-renal fat of mice in the HG group was significantly higher than that of the CG group(P<0.05),and The content of total saturated fatty acid increased significantly(P<0.05),the content of total monounsaturated fatty acid decreased significantly(P<0.05),the content of single polyunsaturated fatty acid in the HG group and the CG group Compared with the significant reduction(P<0.05).The results showed that the perchlorate high-dose group(intake of 10mg/kg bw/d)would affect the fatty acid metabolism disorder in the peri-renal fat of mice,which may lead to the disorder of liver fatty acid metabolism related enzymes and gene regulation in mice,thus affecting fatty acids The synthesis and decomposition of the protein lead to disorder of lipid metabolism in mice.
