Pueraria,as an important plant resource,has been widely used as both a medical and edible raw material in China.Pueraria lobate(Wild.)Ohwi and Pueraria thomsonii Benth are two major species that are used for the preparation of Pueraria products.Accordingly,it is crucial to explore the functional components differences between these two species for the market standardization of Pueraria products.In this study,the content of 11 types flavonoids(puerarin,3’-hydroxy puerarin,3’-methoxy puerarin,daidzin,daidzein,genistein,genistin,ononoside,formononetin,isoliquiritigenin,biochanin A)in Pueraria lobate(Wild.)Ohwi and Pueraria thomsonii Benth in eight main producing areas of Anhui,Sichuan,Shandong,Guangdong,Henan,Jiangsu,Hunan and Shaanxi were detected separately using high performance liquid chromatography-mass spectrometry usage(HPLC-MS).Principal component analysis,cluster analysis,and discriminant analysiswere carried out using SPSS22.0 software.In addition,the capacities of antioxidant from extracts of pueraria were determined by 1,1-diphenyl-2-trinitrophenylhydrazine(DPPH)free radical scavenging testing and 2,2-diazo-bis(3-ethyl-benzothiazole-6-sulfonic acid)the ammonium salt(ABTS)free radical scavenging testing.It was found that the total isoflavone content of Pueraria lobate(Wild.)Ohwi is relatively high in the study.The total isoflavone content in the samples from Shaanxi is the highest,which is 106.11 mg/g.However,the total isoflavone content in the samples from Pueraria thomsonii Benth from Guangdong is the highest,only 3.64mg/g,with significant difference.Among the 11 isoflavones,the content of puerarin is the highest,and the average values of the Pueraria lobate(Wild.)Ohwi samples in Shaanxi and Jiangsu reached 68.58 mg/g and 66.30 mg/g,respectively.The average content of puerarin,daidzin and daidzein in the samples of Guangdong Pueraria thomsonii Benth were the highest values in the eight producing areas,which were2.55 mg/g,0.63 mg/g and 0.32 mg/g,respectively.Principal component analysis and cluster analysis were conducted on samples from different production areas,and the classification results were obviously related to regional characteristics.The main performance is the difference between the south and the north,the coast and the inland.The samples of Pueraria lobate(Wild.)Ohwi from Shaanxi and Jiangsu and the samples of Pueraria thomsonii Benth from Guangdong have significant advantages.ABTS and DPPH free radical scavenging testings show that the pueraria samples have strong antioxidant capacity,and the content of puerarin has an important influence on the antioxidant capacity of pueraria.The free radical scavenging ability of Pueraria lobate(Wild.)Ohwi samples in various regions is Jiangsu> Anhui>Guangdong> Henan> Shandong> Shaanxi> Hunan> Sichuan,while that of Pueraria thomsonii Benth samplesfrom various regions clearance capacity is Jiangsu> Anhui>Sichuan> Henan> Shandong> Hunan> Guangdong> Shaanxi.Pueraria lobate(Wild.)Ohwi has a high medicinal value because its isoflavone content and antioxidant capacity are significantly higher than that of Pueraria thomsonii Benth,while Pueraria thomsonii Benth is more suitable for the development of health food.
